摘要
目的研究环状RNA(circRNA)circ0072995在乳腺癌中的表达及其对乳腺癌细胞增殖、迁移和侵袭的影响。方法收集65例乳腺癌患者癌组织和相应的癌旁组织;用实时荧光定量逆转录聚合酶链反应(qRT-PCR)检测乳腺癌组织和细胞中circ0072995的相对表达水平。向MCF-7细胞中转染circ0072995过表达质粒(circ0072995过表达组)及空质粒(Vector组)以建立过表达circ0072995的体外细胞模型,分别用细胞计数(CCK-8)法和Transwell实验检测过表达circ0072995对乳腺癌细胞株MCF-7细胞增殖、迁移及侵袭的影响。结果circ0072995在乳腺癌癌旁组织和乳腺癌组织中的相对表达水平分别为2.12±0.27,2.89±0.39,差异有统计学意义(P<0.01);circ0072995在乳腺癌细胞系MCF-7、MDA-MB-231和SKBR-3中的相对表达水平分别为2.48±0.29,2.81±0.31,2.89±0.52,均显著高于正常人乳腺上皮细胞系的2.75±0.45(均P<0.01)。与Vector组相比,过表达circ0072995组的光密度值在48,72和96 h明显升高(均P<0.05)。Vector组和过表达circ0072995组的细胞迁移数分别为14.66±1.42,19.33±1.92,细胞侵袭数分别为10.33±1.02,17.33±1.71,差异有统计学意义(P<0.01)。结论circ0072995在乳腺癌组织和细胞中的表达均上调,过表达circ0072995促进了细胞的增殖、迁移及侵袭。
Objective To study the expression of circular RNA(circRNA)circ0072995 in breast cancer and its effects on the proliferation,migration and invasion of breast cancer cells.Methods The cancer tissues and corresponding adjacent tissues of 65 cases of breast cancer were collected.The relative expression level of circ0072995 in breast cancer tissues and cells was detected by real-time quantitative reverse transcription polymerase chain reaction(qRT-PCR).The circ0072995 overexpression plasmid(circ0072995 overexpression group)and the empty plasmid(Vector group)were transfected into MCF-7 cells to establish in vitro cell model of circ0072995 overexpression.The effects of circ-0072995 overexpression on the proliferation,migration and invasion of breast cancer cell line MCF-7 were detected by cell counting kit(CCK-8)and Transwell experiments,respectively.Results The relative expression levels of circ-0072995 in breast cancer adjacent tissues and breast cancer tissueswere 2.12±0.27,2.89±0.39,with significant difference(P<0.01).The relative expression levels of circ_0072995 in breast cancer cell lines MCF-7,MDA-MB-231,and SKBR-3 were 2.48±0.29,2.81±0.31,2.89±0.52,significant higher than that in normal human mammary epithelial cell line MCF-10 A,which was2.75±0.45(P<0.01).Compared with Vector group,the absorbance values of the circ_0072995 overexpressed group were significantly increased at 48,72 and 96 h(all P<0.05).The cell migration numbers of the Vector group and the overexpressed circ_0072995 group were 14.66±1.42,19.33±1.92,the cell invasion numbers were10.33±1.02 and 17.33±1.71,with significant difference(P<0.01).Conclusion The expression of circ_0072995 was up-regulated in both breast cancer tissues and cells,and its overexpression promoted the proliferation,migration and invasion of cells.
