紫茉莉耐盐突变体筛选的初步研究 被引量:2

摘要 以紫茉莉的带节茎段为外植体进行离休培养实验,建立了比较高效的紫茉莉离体再生体系。分析了盐胁迫对紫茉莉不定芽生长的影响,结果表明200 mmol/L NaCl严重抑制了紫茉莉不定芽的生根,故此浓度可作为耐盐突变体的筛选浓度。分析了EMS诱变对紫茉莉不定芽生长的影响,发现经0.8%EMS处理6 h或0.6%EMS处理7 h后,紫茉莉不定芽的致死率在50%左右,因此可以作为紫茉莉突变体筛选的EMS浓度和处理时间。通过合适剂量的EMS诱变和NaCl筛选,初步获得了具有抗性的耐盐紫茉莉突变体植株。 The stem segment with node was used as the explant,and a more efficient in vitro regeneration system of Mirabilis jalapa L.was established.The effect of salt stress on the growth of adventitious buds of Mirabilis jalapa was analyzed.The results showed that 200 mmol/L NaCl severely inhibited the rooting of adventitious buds.Therefore,it was used as the screening concentration for salt-tolerant mutants.The effect of EMS treatment on the growth of the adventitious buds of Mirabilis jalapa was analyzed,and it was shown that 0.8% EMS treatment for 6 h or 0.6% EMS treatment for 7 h was the semi-lethal dose of adventitious buds.Therefore,the concentration and time were used for screening the mutants of Mirabilis jalapa.Through the appropriate dose of EMS mutagenesis and NaCl screening,the salt-tolerant mutants of Mirabilis jalapa were initially obtained.
