多孔四氧化三铁靶向递送尼日利亚菌素诱导细胞焦亡联合光热疗法用于三阴型乳腺癌治疗的研究

摘要 目的探讨多孔四氧化三铁(Fe_(3)O_(4))纳米粒子负载尼日利亚菌素(nigericin)联合光热疗法用于三阴型乳腺癌的治疗效果。方法对合成的多孔Fe_(3)O_(4)纳米粒子进行表征,负载尼日利亚菌素,使用含有200μg/mL的Fe_(3)O_(4)@nigericin诱导4T1细胞焦亡,通过CCK8检测细胞杀伤能力,western blot验证焦亡效果。结果合成的Fe_(3)O_(4)形貌均一,光热转化性能好,负载尼日利亚菌素联合光热治疗具有更强的细胞杀伤能力(P<0.001),诱导的焦亡效果更好(P<0.001)。结论多孔Fe_(3)O_(4)纳米粒子负载尼日利亚菌素联合光热疗法可以通过诱导焦亡对三阴型乳腺癌产生非常好的治疗效果,是三阴型乳腺癌的一种潜在的新型治疗手段。 Objective To investigate the therapeutic effect of nigericin loaded with porous Fe_(3)O_(4) nanoparticles combined with photothermal therapy for triple-negative breast cancer.Methods characterize synthesized porous Fe_(3)O_(4) nanoparticles and loaded with nigericin.The Fe_(3)O_(4)@nigericin nanoparticle with200μg/ml was prepared to induce 4 T1 pyroptosis.The cell killing ability was detected by CCK8,and the pyroptosis effect was verified by Western blot.Results The synthesized Fe_(3)O_(4) showed uniform morphology and good photothermal conversion performance.The combination of loading nigericin and photothermal treatment showed stronger cell killing ability(P<0.001)and better pyroptosis effect(P<0.01).Conclusion Porous Fe_(3)O_(4) nanoparticles loaded with nigericin combining with photothermal therapy can produce very good therapeutic effect for triple-negative breast cancer by inducing pyroptosis,which is a potential new treatment method for triple-negative breast cancer.
