摘要
目的:建立青钱柳中原生苷元槲皮素和山柰素含量测定的方法。方法:样品采用乙醇回流提取,以Symmetry C_(18)柱(250 mm×4.6 mm,5μm)为色谱柱,以乙腈-0.1%磷酸溶液(28∶72)为流动相,流速1.0 ml·min^(-1),进样量10μl,检测波长365nm。结果:槲皮素在0.0150.445μg范围内具有良好的线性关系(r=1.0000),平均回收率为95.3%,RSD为1.31%(n=6);山柰素在0.0190.482μg范围内具有良好的线性关系(r=1.0000),平均回收率为100.8%,RSD为1.29%(n=6)。对收集的11批次青钱柳样品进行了测定,结果槲皮素、山柰素含量范围分别为0.025%0.038%,0.034%0.047%。结论:该方法准确可靠,可作为青钱柳中槲皮素、山柰素的定量分析方法。
Objective:To establish a method for the determination of primary aglycons quercetin and kaempferol in Cyclocarya paliurus.Methods:Samples were extracted by ethanol reflux.A Symmetry C_(18)(250 mm×4.6 mm,5μm)column was adopted with the mobile phase of acetonitrile and 0.1%phosphoric acid solution(28∶78).The flow rate was 1.0 ml·min^(-1) with the injection volume of 10 ul,and the detection wavelength was set at 365 nm.Results:Quercetin had considerable linear relationship within the range of 0.015-0.445μg(r=1.0000),and the average recovery was 95.3%(RSD=1.31%,n=6).Kaempferol had considerable linear relationship within the range of 0.019-0.482μg(r=1.0000),and the average recovery was 100.8%(RSD=1.29%,n=6).Totally 11 batches of Cyclocarya paliurus samples were collected and determined.The content range of primary aglycons quercetin and kaempferol was 0.025%-0.038%and 0.034%-0.047%,respectively.Conclusion:The method is accurate and reliable,which can be used as a quantitative analysis method for primary aglycons quercetin and kaempferol in Cyclocarya paliurus.
