鹅星状病毒感染在河南地区的分子流行病学调查 被引量:5

摘要 鹅星状病毒(goose astrovirus, GAstV)是引起当前雏鹅痛风疾病的主要病原体,其流行严重影响养鹅产业的健康发展。为研究鹅星状病毒在河南地区的流行情况,本研究于2018年从河南12个地市的养鹅场采集了36份雏鹅痛风样品,采用RT-PCR方法进行检测,并对12个地市代表毒株的ORF1b基因进行了序列测定、同源性分析和系统进化分析。研究结果表明,36份样品均为阳性,阳性率为100%;12个地市代表毒株的ORF1b序列长度均为1 551 bp,各自间的核苷酸同源性为98.7%~99.9%,与河南地区的XX、AstV/HN01/Goose/0103/18、AstV/HB01/Goose/0123/19、AstV/AH01/Goose/0512/18等代表毒株核苷酸同源性为97.9%~99.5%,与国内其他地区的GD、AstV/SDPY/Goose/1116/17、AAstV/Goose/CHN/2017/SD01等代表毒株核苷酸同源性为98.4%~99.7%;系统进化分析显示,12个地市代表毒株与河南及国内其他地区代表毒株,在进化关系上处于同一分支,属于禽星状病毒1群。本研究明确了河南地区鹅星状病毒的分子流行情况,并为鹅痛风病的防治提供了线索和思路。 Goose astrovirus(GAstV) is the agent causing the current gosling gout epidemic, which seriously affects the healthy development of China’s goose industry. In order to understand the prevalence of GAstVs in the Henan area, 36 samples collected from goslings suffering from gout were collected from goose farms in 12 regions in the Henan area in 2018.Then, RT-PCR was used to detect the presence of GAstV in the samples. And homological analysis and phylogenetic analysis were conducted based on the ORF1 b genes of the representative strains from the 12 regions. The results showed that the 36 samples were all positive with GAstV and the length of the ORF1 b genes of the 12 representative strains all reached 1 551 bp. The nucleotide homology within the 12 strains was 98. 7%-99. 9% and the nucleotide homology of the 12 strains with other representative strains circulating in the Henan area,including XX,AstV/HN01/Goose/0103/18,AstV/HB01/Goose/0123/19,and AstV/AH01/Goose/0512/18,was 97. 9%-99. 5%. The nucleotide homology of the 12 strains with the representative strains in the other regions in China,such as GD,AstV/SDPY/Goose/1116/17,and AAstV/Goose/CHN/2017/SD01,was 98. 4%-99. 7%. Phylogenetic analysis showed that the 12 representative strains were in the same branch with that of the strains prevalent in the other parts of the Henan area and the other regions in China,and they were classified as avastrovirus group 1. This study revealed the molecular epidemiology of the GAstV s currently prevalent in the Henan area,and provided clues and ideas for the prevention and treatment of the disease.
