摘要
运用响应面分析法对高压脉冲电场提取橘皮中黄酮的工艺进行了优化。在单因素试验的基础上,以乙醇体积分数、m(乙醇)∶m(橘皮渣)、电场强度和脉冲数为自变量,橘皮总黄酮得率为响应值,采用响应面分析法建立高压脉冲电场提取橘皮总黄酮的回归方程,优化并最终确定提取黄酮的最优工艺为:乙醇体积分数为75%、m(体积分数为75%乙醇)∶m(橘皮渣)为36∶1、电场强度19 kV/cm、脉冲数10个,总黄酮得率为2.312%(与理论预测值比较仅相差1.11%)。验证试验值与模型预测值拟合良好,模型对于高压脉冲电场提取橘皮渣总黄酮的工艺设计具有指导价值。
The response surface method(RSM) was used to optimize the process for total flavones of orange peel by the high-voltage pulsed electric field. On the basis of single factor experiment with ethanol concentration, mass ratio of ethanol to orange peel, electric field intensity and pulse number as independent variables and the yield of orange peel total flavonoids as the response value, RSM was used to establish a regression equation for the extraction of flavonoids from orange peel by the high-voltage pulsed electric field. The optimal process for extracting flavonoids was determined as follows: volume fraction of ethanol was 75%, mass ratio of ethanol to orange peel was 36∶1(m∶m), electric field intensity was 19 kV/cm, pulse number was 10 so that total flavonoid yield can reach 2.312%. In comparison with the theoretical prediction value, the difference was only 1.11%. It is verified that the test value fits well with the predicted value of the model, and the model has guiding value for the process design of high-voltage pulsed electric field extraction of flavonoids from orange peel.
