高效液相色谱法快速测定保健食品中叶黄素含量研究

Research on Rapid Determination of Lutein Content in Health Food by High Performance Liquid Chromatography

本文研究了快速提取保健食品中叶黄素并进行含量分析的高效液相色谱法。样品经皂化处理后定容,直接过膜上机,以甲醇和甲基叔丁基醚为流动相,用C30色谱柱进行梯度洗脱,在445nm波长下检测,外标法定量。叶黄素在0.02~2.0μg/mL范围其线性相关系数大于0.9999,检出限为10μg/100g,定量限为40μg/100g,回收率在88.7%~105.0%之间,相对标准偏差小于3.5%。提取过程简单快速,适合用于保健食品中叶黄素含量的测定,测定结果准确。

A high performance liquid chromatography(HPLC)method for rapid extraction and content analysis of lutein in health food was studied.After saponification,the sample was put to a constant volume and directly passed through the membrane.Methanol and methyltertbutylether were used as mobile phases.C30 column was used for gradient elution.The sample was detected at 445 nm and quantified by external standard method.The linear correlation coefficient of lutein was greater than 0.9999 in the range of 0.02-2.0μg/mL.the detection limit was 10μg/100g and the limit of quantification was 40μg/100g.The recoveries ranged from 88.7%to 105.0%,and the relative standard deviation was less than 3.5%.The extraction process is simple and rapid,which is suitable for the determination of lutein in health food with accurate results.
