响应面优化药桑椹多糖超声提取工艺及其抗氧化活性研究 被引量:29

Optimization of ultrasonic extraction of polysaccharides from medicinal mulberry by response surface methodology and its antioxidant activity

摘要 为研究药桑椹多糖的超声提取工艺及体外抗氧化活性。以多糖得率为指标,采用单因素试验结合Box-Behnken响应面法得到最优的药桑椹多糖提取工艺。通过检测1,1-二苯基-2-三硝基苯肼(DPPH)自由基,2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS)自由基,羟基自由基清除活性,铁离子还原/抗氧化能力(FRAP)对药桑椹多糖的体外抗氧化活性进行研究。结果表明药桑椹多糖的最佳提取工艺为:超声功率100W,提取时间35min,液料比27mL/g,提取次数3次。在最优条件下,多糖得率为10.81%。体外抗氧化研究表明在质量浓度0.2~1.2mg/mL范围内,药桑椹多糖具有较强的抗氧化活性,其清除DPPH、ABTS、羟基自由基的最大清除率分别为71.21%、38.52%、62.57%,其在FRAP试验中,FRAP最大值为28.44。本研究为进一步研究药桑的药用价值提供了理论依据。 In order to study ultrasonic extraction technology and in vitro antioxidant activity of Polysaccharides from medicinal mulberry,yield of polysaccharides was selected as the index.The single factor test combined with Box-Behnken response surface method was used to obtain the optimal extraction process.The antioxidant activity of mulberry polysaccharide in vitro was studied by DPPH,ABTS,hydroxyl radical and FRAP tests.The results showed that the optimum extraction conditions were:ultrasonic power 100W,extraction time 35min,liquid-solid ratio 27mL/g,and extraction 3 times.Under the optimal conditions,the yield was 10.81%.In vitro studies showed that the mulberry polysaccharide had strong antioxidant activity in the range of 0.2~1.2mg/mL.the maximum scavenging rates of DPPH,ABTS and hydroxyl radical were 71.21%,38.52% and 62.57%,respectively.In FRAP test,the maximum FRAP value was 28.44.This study provides a theoretical basis for further research of medicinal value of medicinal mulberry in the future.
