摘要
目的探讨组蛋白去甲基化酶JMJD2B和HIF-1α在横纹肌肉瘤(RMS)组织中的表达及意义。方法选取78例RMS患者通过随访获取生存时间,采用免疫组织化学染色法检测JMJD2B和HIF-1α在RMS组织中的表达情况并分析其与患者临床病理特征及预后的关系。结果78例样本中,JMJD2B和HIF-1α表达阳性例数分别为46例(59.0%)和44例(56.4%)。JMJD2B和HIF-1α的表达成正相关(P<0.001,r=0.518)且与肿瘤的危险度、治疗、转移、分期有关(均P<0.05)。生存分析显示危险度的级别、新辅助化疗的有无、HIF-1α和JMJD2B表达的高低与患者预后相关(均P<0.05)。多因素Cox回归分析进一步显示JMJD2B(HR=3.161,P=0.039)和危险度(HR=2.925,P=0.001)可作为RMS患者独立的预后因素。结论JMJD2B和HIF-1α的表达与横纹肌肉瘤的总体生存密切相关,JMJD2B是横纹肌肉瘤患者的独立预后指标。
Objective To explore the expression and prognostic significance of histone demethylase JMJD2B and HIF-1αin rhabdomyosarcoma(RMS)tissues.Methods A total of 78 RMS patients were followed up to obtain survival time,and immunohistochemical staining was used to detect the expression of JMJD2B and HIF-1αin RMS tissues and analyze their relation with clinicopathological characteristics and prognosis.Results Among the 78 samples,there were 46(59.0%)and 44(56.4%)cases of positive expression of JMJD2B and HIF-1α,respectively.The expression of JMJD2B and HIF-1αwere positively correlated(P<0.001,r=0.518)and related to tumor risk,metastasis,IRS staging and neoadjuvant chemotherapy(all P<0.05).The Kaplan-Meier survival curve and univariate Cox regression analysis showed that the risk level,neoadjuvant chemotherapy,the expression of HIF-1αand JMJD2B were related to the prognosis of patients(all P<0.05).Multivariate Cox regression analysis showed that JMJD2B(HR=3.161,P=0.039)and risk degree(HR=2.925,P=0.001)could be used as independent prognostic factors for RMS patients.Conclusion JMJD2B and HIF-1αexpression are both significantly related to the overall survival of rhabdomyosarcoma patients,and JMJD2B is an independent prognostic indicator.
