胚蛋给养二糖和蛋氨酸对鹅肠道发育的影响

摘要 试验研究胚蛋给养二糖和蛋氨酸对鹅肠道发育的影响。选择发育良好吉林白鹅胚蛋600枚,分为4组,每组3个重复,每个重复50枚胚蛋。孵化第24 d进行胚蛋给养,每枚胚蛋注射1.5 mL给养液,对照组不进行给养,根据体重每个处理随机分为3个重复,每个重复30只雏鹅进行饲养,饲养期为28 d。于孵化第27 d(E27)、出雏当天(0 d)、1、4、7、14、21、28 d采集样品,各组选取胚蛋或健康鹅雏各6枚/羽。采集空肠组织样(n=6)。测定空肠绒毛面积、空肠二糖酶和谷胱甘肽过氧化物酶活性、空肠SI、SGLT1和GLUT2 m RNA相对表达量。结果显示,从E27到4 d,胚蛋给养蛋氨酸可以显著增加雏鹅小肠指数。胚蛋给养二糖和蛋氨酸对E27雏鹅小肠指数增加有显著性交互作用。胚蛋给养二糖能通过提高小肠绒毛表面积、提高二糖酶活性和提高SGLT1、GLUT2和SI mRNA相对表达量来促进肠道的发育和对营养物的消化与吸收能力。胚蛋给养蛋氨酸可以通过提高肠道谷胱甘肽过氧化物酶活性来增强肠道的氧化还原能力,促进肠道的发育,胚蛋给养二糖和蛋氨酸对空肠绒毛面积的增加和GLUT2 mRNA的表达量的增高有显著的交互作用。 The experiment studied the effect of disaccharides and methionine from embryo eggs on goose intestinal development. Selected 600 goose eggs containing viable embryos and divided into four groups with three replicates in each group and 50 eggs in each replicate. 1.5 mL solution was injected into each egg on the 24 th days of incubation(E24).Nothing was injected into the control group. Goslings in each treatment were randomly divided into three replicates with30 goslings for each replicate and the feeding period was 28 d. On the E27, the hatching day(0 d), the 1, 4, 7, 14, 21, 28 d post-hatch respectively, jejunum within each group were sampled(n=6) for morphological analysis, and mucosa enzyme activities, antioxidant ability and SI, SGLT1 and GLUT2 mRNA relative expression were determined. The experiment indicates that: from E27 to 4 d, in ovo feeding Met can significantly increased the mass index of small intestine of goslings. In ovo feeding DS and Met had a significant interaction effect on the increase of small intestinal mass index of E27 goslings. In ovo feeding DS can promote intestinal development and improve the ability of the digestion and absorption in small intestine due to the increase of villus area of small intestinal, the enhancement of disaccharidase activity and the improvement of SGLT1, GLUT2 and SI mRNA relative expression. The experiment indicates that in ovo feeding Met reduced cell apoptosis by improving GSH-Px activity to enhance antioxidant ability of small intestine. There were significant interactive effects of DS and Met on improvement of villus area of small intestinal and GLUT2 mRNA relative expression.
