摘要 目的:评价荷叶水提取物的急性毒性及辅助降血脂功效,为荷叶水提取物在功能食品及药品中的开发利用提供依据。方法:通过大鼠急性经口毒性试验考察其安全性;通过考察荷叶水提取物对高脂血症模型大鼠体重、血脂、体脂、肝脏,以及肝脂肪变性HepG2细胞脂质代谢的影响,评价其辅助降血脂功效。结果:雄性大鼠LD50为7.5 g·kg^(-1) BW,雌性大鼠LD50为6.1 g·kg^(-1) BW,平均LD50为6.7 g·kg^(-1) BW,安全性良好。与模型对照组相比,各剂量荷叶水提取物大鼠的血清甘油三酯(TG)水平明显降低,高、低剂量组大鼠的血清总胆固醇(TC)水平均明显降低。此外,中、低剂量组大鼠1~4周体重、空腹体重降低,中剂量组大鼠肝脏重量降低。荷叶水提取物对肝脂肪变性HepG2细胞内脂质含量影响不显著。结论:荷叶水提取物安全性较高,为实际无毒级,具有较好的辅助降血脂功效,可以广泛应用于降脂产品中。 Objective:To evaluate the acute toxicity and auxiliary hypolipidemic activity of lotus leaf aqueous extract(LLAE),which would provide experimental basis for the application and product development of LLAE.Methods:The safety of LLAE was assessed by acute oral toxicity test on rats.The auxiliary hypolipidemic activity was evaluated by investigating the effect of LLAE on the body weight,blood lipid,body fat,liver in hyperlipidemia model rats,and lipid metabolism of HepG2 cell model.Results:Median lethal dose(LD50)was 7.5 g·kg^(-1) BW for male,6.1 g·kg^(-1) BW for female and 6.7 g·kg^(-1) BW in total.Compare with the model group,each dose group of LLAE and positive group could significantly reduce the content of serum triglyceride(TG)in rat,meanwhile,the content of serum total cholesterol(TC)in rat were significantly decreased both in high and low dose groups with positive group. In addition, the body weight and fasting weight of rats in middle and low dose groups were decreased during 1~4 weeks. The liver weight of rats in middle dose group was decreased. The LLAE showed no significant effect on lipid content in HepG2 cells with hepatic steatosis. Conclusions: LLAE is safe and actually non-toxic and has the auxiliary hypolipidemic activity. It could be widely used in lipid-lowering products. but no significant effect on lipid metabolism of hepatic steatosis HepG2 cells. Its hypolipidemic effect may work through other pathways.
