摘要
目的探讨肉苁蓉苯乙醇总苷(CPhGs)对H22荷瘤小鼠皮下移植瘤的抑制作用。方法利用H22肝癌细胞株建立荷瘤小鼠模型,60只雄性昆明小鼠随机分为6组(10只/组):正常对照组(蒸馏水)、模型组(蒸馏水)、模型+阳性对照组(西黄丸780 mg/kg·bw)、模型+CPhGs低剂量组(125 mg/kg·bw)、模型+CPhGs中剂量组(250 mg/kg·bw)和模型+CPhGs高剂量组(500 mg/kg·bw)。除正常对照组以外,其余各组造模后24 h,每组均按设定剂量灌胃给予蒸馏水或受试药物CPhGs、西黄丸,1次/d,连续20 d。比较各组小鼠脏器指数和瘤体体积、瘤指数、抑瘤率,血清中AST、ALT活力,AFP、TNF-α含量。结果CPhGs高剂量组瘤体质量、瘤指数和瘤体体积低于模型组,差异具有统计学意义(P<0.05);与阳性对照组相比,CPhGs高剂量组抑瘤率较高,差异具有统计学意义(P<0.01)。与模型组比较血清中AST与ALT活力,CPhGs高剂量组活力降低,差异具有统计学意义(P<0.05);与正常对照组比较血清中AFP含量,模型组与各干预组含量均升高,差异具有统计学意义(P<0.01);CPhGs高剂量组血清中TNF-α含量低于模型组,差异具有统计学意义(P<0.05)。结论一定剂量的CPhGs可抑制H22荷瘤小鼠皮下移植瘤生长。
Objective To investigate the anti-tumor effect of phenylethanol glycosides from Cistanche deserticola(CPhGs)on H22 tumor-bearing mice.Methods H22 hepatoma mice models were established.Sixty male Kunming mice were randomly divided into six groups(10 mice/group):normal control group(distilled water),model group(distilled water),model+positive control group(Xihuang Wan 780 mg/kg·bw),model+CPhGs low dose group(125 mg/kg·bw),model+CPhGs medium dose group(250 mg/kg·bw),model+CPhGs high dose group(500 mg/kg·bw).Except for the normal control group,the rest of the groups were given distilled water or the test drug CPhGs and Xihuang Wan by gavage at the set dose on the second day after modeling,once a day for 20 consecutive days.The organ index,tumor volume,tumor index,tumor inhibition rate,serum AST,ALT activity,AFP and TNF-αcontent in each group were compared.Results The tumor mass,tumor index,and tumor volume of the CPhGs high-dose group were lower than those of the model group,and the difference was statistically significant(P<0.05).Compared with the positive control group,the high-dose CPhGs group had a higher tumor inhibition rate,and the difference was statistically significant(P<0.01).Compared with the model group,the CPhGs high-dose group had lower serum AST and ALT activity,and the difference was statistically significant(P<0.05).Compared with the normal control group,the serum AFP content was increased in the other 5 groups,and the difference was statistically significant(P<0.01).The serum TNF-αcontent of CPhGs high-dose group was lower than that of model group,and the difference was statistically significant(P<0.05).Conclusion CPhGs can inhibit tumor growth in H22 tumor-bearing mice.
