摘要
目的探讨不同年龄小鼠脾脏B细胞表面免疫球蛋白分子、相关表面分子的表达和功能差异。方法不同年龄小鼠的脾脏制备成单个核细胞后进行流式细胞术染色,分析B细胞表面分子的表达。纯化CD19+B细胞,用LPS或CpG刺激培养,检测细胞因子和抗体的分泌。结果小鼠脾脏中的B细胞、浆母细胞和浆细胞数随小鼠年龄增加呈升高趋势,且B细胞表面免疫球蛋白和多种分子的表达也随小鼠年龄的增加而不同。相同刺激条件下,相比新生小鼠,成年小鼠脾脏B细胞产生细胞因子能力较弱,产生抗体能力更强。结论小鼠脾脏B细胞的表型和功能随年龄的变化而不同。本研究揭示了小鼠发育过程中B细胞的表型及功能改变,为B细胞在不同年龄段的动物及人群中的分布和功能研究奠定了基础。
This study designed to investigate the expression of immunoglobulins(Igs)and other surface molecules on splenic B cells,and the functional differences in naïve mice at different ages.Splenocytes from mice were isolated and then stained.CD19+B cells were purified and cultured with LPS or CpG for detecting the production of cytokines and antibodies.Data indicated that the numbers of splenic B cells,plasmablasts and plasma cells had increasing trends with the age,and various surface markers on splenic B cells were different.Under the same stimulation conditions,compared with newborn mice,the production of cytokines was lower and the antibodies secretion were much higher on splenic B cells of adult mice.The phenotypes and functions of splenic B cells in mice were gradually changed with age.Our study elucidated the phenotypic and functional changes of B cells during the development of mice,which lay the foundation for the study on the distribution and functional differences of B cell in animals and people at different ages.
