超微肿痛贴对膝骨关节炎模型兔关节液IL-1β、IL-6及关节软骨MMP-3、Ⅱ型胶原蛋白表达的影响 被引量:7

摘要 目的:观察超微肿痛贴贴敷治疗对膝骨关节炎(KOA)模型兔关节液IL-1β、IL-6及关节软骨MMP-3、Ⅱ型胶原蛋白表达的影响。方法:将50只3月龄新西兰兔随机分为空白组(12只)和造模组(38只),造模组进行造模后随机分为模型组、氟比洛芬巴布膏组、超微肿痛贴组,每组12只。空白组及模型组不做任何干预,超微肿痛贴组、氟比洛芬巴布膏组分别给予超微肿痛贴及氟比洛芬巴布膏治疗。干预4周后,取兔膝关节液用ELISA法测定IL-1β、IL-6水平,并取膝关节行HE染色观察病理形态学改变,取膝关节软骨采用Western blotting检测软骨中MMP-3、Ⅱ型胶原蛋白表达量。结果:HE染色后光镜下发现超微肿痛贴组兔膝关节软骨损伤程度较模型组减轻,Mankin’s评分降低。超微肿痛贴组及氟比洛芬巴布膏组兔关节液中IL-1β、IL-6的水平和关节软骨中MMP-3的表达明显低于模型组(P<0.05),Ⅱ型胶原蛋白表达明显高于模型组(P<0.05)。结论:超微肿痛贴能够有效减轻兔膝骨关节炎的软骨损伤,抑制MMP-3表达,上调Ⅱ型胶原蛋白表达,减少关节液中IL-1β及IL-6水平,这可能为其治疗膝骨关节炎的部分作用机制。 Objective:To observe the effect of Chaowei Zhongtong Patch on the expression of IL-1βand IL-6 in the synovial fluid and the expression of MMP-3 and typeⅡcollagen in articular cartilageof rabbits with knee osteoarthritis(KOA).Methods:A total of 503-month-old New Zealand rabbits were randomly divided into blank group(12 rabbits)and model group(38 rabbits).After model-building,they were randomly divided into model group,flurbiprofen cataplasm group,Chaowei Zhongtong Patch group,with 12 rabbits in each group.The blank group and the model group did not do any intervention.The Chaowei Zhongtong Patch group and flurbiprofen cataplasm group were given corresponding drug treatments.After the 4-week intervention,the levels of IL-1βand IL-6 in rabbit knee joint fluid were measured by ELISA,and the pathological changes of knee joints were observed by HE staining.Besides,the expression levels of MMP-3 and collagen II in articular cartilage were detected by Western blotting.Results:After HE staining,it was found under light microscope that the degree of cartilage damage in Chaowei Zhongtong Patch group was less than that of the model group,and the Mankin’s score was lower.The levels of IL-1βand IL-6 in the synovial fluid and the expression of MMP-3 in the articular cartilage of the rabbits in Chaowei Zhongtong Patch group and flurbiprofen cataplasm group were significantly lower than those in the model group(P<0.05),and the expression of typeⅡcollagen was significantly higher than that of the model group(P<0.05).Conclusion:Chaowei Zhongtong Patch can effectively reduce the cartilage damage of rabbit knee osteoarthritis,inhibit the expression of MMP-3,up-regulate the expression of typeⅡcollagen,and reduce the levels of IL-1βand IL-6 in the synovial fluid,which may be part of the mechanism of its treatment of knee osteoarthritis.
