补肾解毒方对辐射致小鼠胎盘结构损伤的防护作用研究 被引量:1

摘要 目的探究中、低剂量辐射对小鼠胎盘结构的影响及补肾解毒方对胎盘损伤的防护作用。方法选取受孕后孕鼠25只,随机分为5组,分别为空白对照组,1 Gy辐射组,4 Gy辐射组,1 Gy辐射+中药组,4 Gy辐射+中药组,自受孕第1天开始每日分别予1 Gy辐射组,4 Gy辐射组组生理盐水灌胃,1 Gy辐射+中药组,4 Gy辐射+中药组补肾解毒方溶液灌胃,10 d后分别予1 Gy、4 Gy 60 Co-γ射线照射,照射后仍以以上方法灌胃10 d,剖腹取出仔鼠及胎盘,称重胎盘,电镜观察胎盘切片。结果与空白对照组相比,1 Gy辐射组和1 Gy辐射+中药组仔鼠胎盘组织损伤轻微,胎盘重量及胎盘结构改变均不明显;4 Gy辐射组胎盘重量较空白对照组明显降低(P<0.05),胎盘结构疏松紊乱,主要可见血管管腔扩大,内有血凝块,血管内红细胞明显减少,组织间有淤血现象,纤维素样坏死等表现;4 Gy辐射+中药组胎盘重量较空白对照组亦明显降低(P<0.05),但较单纯4 Gy辐射组有所增加(P<0.05),胎盘血管组织结构尚清晰,血管扩张不明显。结论中高剂量辐射会导致胎盘重量减低和组织损伤,尤其血管损伤明显,补肾解毒方对中高剂量辐射具有较好的防护作用。 Objective To investigate the effect of moderate and low dose radiation on placenta structure of mice and the protective effect of Bushen Jiedu Recipe(补肾解毒方) on placental injury.Methods Twenty-five pregnant mice were randomly divided into 5 groups,which were normal group,1 Gy radiation group,4 Gy radiation group,1 Gy radiation+traditional Chinese medicine group,4 Gy radiation+traditional Chinese medicine group.From the 1st day of conception,normal saline was administered to the 1 Gy radiation group,4 Gy radiation group daily respectively.Bushen Jiedu Recipe solution was administered to the 1 Gy radiation+traditional Chinese medicine group,4 Gy radiation+traditional Chinese medicine group.10 days later,1 Gy and 4 Gy 60 Co-γ were given to each group respectively.After radiation,the above gavage method was continue to administer for 10 days.The baby mice and placenta were removed by laparotomy.We weighed the placenta and observed the placental sections by electron microscopy.Results Compared with the normal group,the placental tissue damage of 1 Gy radiation group and 1 Gy radiation+traditional Chinese medicine group placental was slight,and the placental weight and the placenta structure were not significantly changed.The placental weight of the 4 Gy radiation group was significantly lower than that of the normal group(P<0.05).The structure of placenta was loose and disordered,mainly with enlarged vascular lumen,blood clots,significantly reduced red blood cells,congestion between the tissues,and fibrinoid necrosis.The placental weight of the 4 Gy radiation+traditional Chinese medicine group was also significantly lower than that of the normal group(P<0.05),but the vascular structure of placenta was still clear,and the vascular dilatation was not obvious compared with that of the 4 Gy radiation group.Conclusion Moderate and low dose radiation can cause placenta weight loss and tissue damage,especially blood vessel damage.Bushen Jiedu Recipe has better protective effect on middle and high dose radiation.
