酒糟泥中纤维素降解菌的筛选与鉴定 被引量:1

摘要 利用羧甲基纤维素钠培养基及刚果红染色从酒糟泥中筛选具有纤维素降解能力的菌株,利用DNS法测定纤维素降解能力最强的菌株6 d内的滤纸酶活,并从形态学和分子生物学两方面对该菌株进行初步鉴定。结果表明:从酒糟泥中获得4株长势良好的菌株,其中菌株FIB-3刚果红染色后的透明圈直径与菌落直径比值(1.88±0.175)最大,并且该菌株在第5 d时滤纸酶活达到最高(8.436 U/mL);结合形态学观察和分子生物学分析,初步确定菌株FIB-3为一株贝莱斯芽孢杆菌。 In order to screen cellulose-degrading strains from liquor sludge,sodium carboxymethyl cellulose medium and Congo red staining were used,the filter paper enzyme activity of the best cellulose-degrading strain in 6 days was determined by DNS method,and that strain was identified based on morphological characteristics and molecular biology analysis.Results showed that among the four strains isolated from wine sludge,there was only one strain FIB-3 that could produce the largest ra-tio(1.88±0.175)of the diameter of transparent circle to that of the colony after Congo red staining.And the filter paper enzyme activity of strain FIB-3 reached the highest(8.436 U/mL)on the fifth day.According to the morphological observation and mo-lecular biology analysis strain FIB-3 was preliminarily identified as Bacillus velezensis.
