Expression and significance of LASP-1 in renal cell carcinoma

摘要 目的观察LASP-1在肾细胞癌中的表达,探讨其在肾癌发生发展中的作用。方法应用免疫组织化学和Western blot检测2015年5月至2019年5月徐州医科大学附属市立医院82例肾癌患者手术切除的癌组织及癌旁组织中的LASP-1表达结果LASP-1在肾细胞癌组织中的阳性表达率为92.68%(76/82),其中强阳性为29.27%(24/82);癌旁组织阳性表达为26.83%(22/82),两者比较差异有统计学意义(P<0.01)£Western blot检测发现,癌旁组织LASP-1的表达量(0.818±0.064)和无淋巴结转移组LASP-1的表达量(1.443±0.154)明显低于有淋巴结转移癌组织的表达量(2.030±0.183),差异有统计学意义(P><0.05)。LASP-1的表达与TNM分期(rs=0.386)和有无淋巴结转移(rs=-0.711)有相关性(P<0.05),但是与肿瘤大小(rs=-0.172)及Furhman分级(H=7.235)无相关性(P>0.05)。结论检测肿瘤内LASP-1表达有助于判断肾细胞癌侵袭性和估计预后。 Objective To observe the expression and significance of LASP-1 protein in renal cell carcinoma(RCC).Methods Immunohistochemistry and Western blot were used to detect the LASP-1 expression in 82 cases of RCC tissues and normal tissues from May 2015 to May 2019 in the affiliated Xuzhou municipal hospital of Xuzhou medical university.Results We found that LASP-1 was highly expressed in RCC.The positive rate of LASP-1 expression in RCC was 92.68%(strongly positive expression rate of 29.27%),which was significantly higher than that in the adjacent tissue(26.83%,P<0.01).The expression level was positively correlated with tissue type,TNM stage and lymph node metastasis(P<0.05),hut not with tumor size or Furhman grade(P>0.05).We extracted the tissue protein of fresh RCC after resection and detected it by western blot in order to further investigate the correlation between LASP-1 expression and lymph node metastasis in RCC.Western blot analysis showed that the expression level of LASP-1 in adjacent tissues(0.818±0.064)and in the group without lymph node metastasis(1.443±0.154)was significantly lower than that in the group with lymph node metastasis(2.030±0.183),with statistical significance(P<0.05).The expression of LASP-1 was correlated with tissue type,TNM stage(rs=0.386,P<0.05)and lymph node metastasis(rs=-0.711,P<0.05).However,there was no correlation with tumor size(rs=-0.172,P<0.05)or Furhman grade(H=1.235,P>0.05).Conclusions Intratumor LASP-1 testing can help determine tumor invasiveness and predict the prognosis of RCC.
