摘要
转录因子作为转录水平基因表达调控的重要上游因子,广泛参与玉米生长发育相关基因的表达调控,而目前关于转录因子候选基因关联分析的报道较少。为了研究转录因子对玉米表型的潜在影响,本研究以508份玉米自交系组成的关联群体全基因组范围内属于81个转录因子家族的2034个转录因子序列,结合21种已公布的玉米农艺及产量性状表型数据,进行了候选基因关联分析。结果表明,在Q+K模型下,共鉴定到3578个SNP与21种表型显著关联(P≤10^(-3))。这些SNP解释表型变异范围为2.37%-68.15%。其中轴色显著关联的转录因子最多,为105个;ZmMADS10关联到的表型最少,只有8个。在家族成员10个以上的转录因子家族中,G2-like家族转录因子显著性成员比例最高,FHA家族平均每个基因含有的SNP个数最多。这些显著SNP位于内含子区、外显子区、5'UTR区和3'UTR,分别占比47.19%、28.20%、12.23%和12.39%。本研究捕获了对玉米生长发育存在广泛调控的转录因子SNP,为玉米分子标记辅助育种提供了基因资源和分子标记。
Transcription factors are key factors involved in gene expression regulation.They participate in maize growth and tissue development.However,the genome wide association analysis(GWAS)of maize transcription factors has not yet been systemically described.In order to study the potential effect of transcription factors on maize phenotypes,508 maize inbred lines were served as the a natural GWAS population.Based on 21 kinds of maize major agronomic and quality traits,GWAS were performed on 2034 maize transcription factors.The result showed that,under Q+K model,3578 SNPs were detected and significantly associated with one of all the traits at least(P≤10-3).Each SNP could explain phenotype variation from 2.37%to 68.15%.About 105 TF genes were significant associated with the trait of cob color,which was most abundant.And,all SNPs in one gene,ZmMADS10,associated with eight different kinds of phenotypes,mostly.Among transcription factor families containing more than 10 members,G2-like family accounted for the largest proportion of the genes significantly associated with traits,wherever FHA family contained the most average SNP members for each TF genes.It was found that SNPs located in the gene region of intron,exon,5’UTR and 3’UTR,accounting for 47.19%,28.20%,12.23%and 12.39%,respectively.These results show that,transcription factors participate in regulation of gene expression during maize growth and development.In addition,our study analyzed the associated SNPs,which would help to provide gene resources and molecular markers for further molecular marker assisted breeding.
