外泌体circRNA在肝癌中的作用及其分子机制研究 被引量:2

摘要 目的探讨外泌体circRNA在肝癌中的作用及其可能的分子机制。方法收集20例原发性肝癌患者的肿瘤组织及癌旁组织标本,通过基因芯片比较肿瘤组织与癌旁组织中circRNA的表达。针对差异表达的circ-CDYL,提取外泌体并检测其中circ-CDYL的表达水平;将circ-CDYL高表达和低表达的细胞外泌体与肝癌细胞进行共培养,检测circ-CDYL对肝癌细胞增殖、侵袭能力的影响。结果肿瘤组织与癌旁组织中差异表达最显著的circRNA为circ-CDYL;稳定过表达circ-CDYL的细胞外泌体中circ-CDYL的含量显著较高,稳定干扰的表达则较少;与circ-CDYL高表达的细胞外泌体相比,circ-CDYL低表达的细胞外泌体与肝癌细胞共培养后,细胞增殖和侵袭受到抑制。结论circ-CDYL可促进肝癌细胞的增殖和转移。 Objective To explore the role of exosome circRNA in liver cancer and its possible molecular mechanism.Methods The tumor tissues and paracancerous tissues of 20 patients with primary liver cancer were collected,and the expressions of circRNA in tumor tissues and paracancerous tissues were compared by gene microarray.For the differential expression of circ-CDYL,the exosomes were extracted and the expression level of circ-CDYL was detected.The exosomes with high and low expression of circ-CDYL were co-cultured with hepatoma cells to detect the impact of circ-CDYL on the proliferation and invasion of hepatoma cells.Results The most differentially expressed circRNA in tumor tissues and paracancerous tissues was circ-CDYL;The content of circ-CDYL in exosomes with stable overexpression of circ-CDYL was significantly higher,but the expression of stable interference was less;Compared with exosomes with high expression of circ-CDYL,the proliferation and invasion of hepatoma cells co-cultured with exosomes with low expression of circ-CDYL were inhibited.Conclusions circ-CDYL can promote the proliferation and metastasis of hepatoma cells.
