摘要 目的探讨AD病人血清及尿液中的代谢物变化,为AD的早期诊断提供依据。方法应用氨基酸自动分析仪测定AD老人(AD组)和健康老人(对照组)血液及尿液中17种氨基酸的含量。采用超高效液相色谱飞行时间质谱仪对所有尿液样本进行全成分检测,利用多变量统计分析筛选生物标志物。结果尿液中苏氨酸、丝氨酸、缬氨酸和赖氨酸的含量在AD组和对照组间差异具有统计学意义(P<0.05);血清中苏氨酸、半胱氨酸、蛋氨酸和亮氨酸的含量在AD组和对照组间差异具有统计学意义(P<0.05)。AD组病人尿液经过筛选检测出15种代谢产物,而对照组中未检出。结论健康老人与AD老人的血清和尿液的代谢物含量具有明显差异,部分代谢产物可作为潜在的代谢标志物用于AD的临床诊治。 Objective To explore the changes of metabolites in serum and urine in the patients with Alzheimer's disease(AD),so as to provide basis for early diagnosis of AD.Methods The content of 17 kinds of amino acids in blood and urine of the elderly AD patients(AD group)and the healthy elderly(control group)were detected by automatic amino acid analyzer.All urine samples were detected by UPLC-QETOF-MS,and biomarkers were screened by multivariate statistical analysis.Results The levels of the amino acids as Thr,Ser,Val and Lys in urine and the levels of Thr,Cys,Met and Leu in blood were significantly different between AD group and control group(P<0.05).There were 15 metabolites in urine of AD group were detected,which were not detected in control group.Conclusions Compared with the healthy elderly,there are significant differences in the contents of metabolites in serum and urine of the elderly with AD.Some metabolites may be used as potential metabolic markers for clinical diagnosis and treatment of AD.
