不同强度流体剪切应力作用下成骨细胞的形态学改变 被引量:1

摘要 目的:研究不同强度流体剪切应力对人成骨肉瘤细胞MG-63的形态学影响。方法:运用定常流加载系统对细胞玻片加力,观察分析流体剪切应力7dynes/cm_(2)和12 dynes/cm_(2)作用15、30、60和120 min时,力学信号对成骨细胞形态的影响;用苏木精-伊红染色和倒置相差显微镜观察成像法展示细胞形态学的改变。结果:作用时间不超过60 min时,两种强度流体剪切应力均使细胞极性增强,排列更规则;作用时间达120 min时细胞形态变得不规则,异常核分裂相增多。细胞平均面积测定结果显示强度7 dynes/cm_(2)的剪切应力作用30、60和120 min时细胞面积显著大于对照组(P<0.05);强度12 dynes/cm_(2)的剪切应力作用60 min和120 min时细胞面积显著大于对照组(P<0.05);在不同剪切应力强度相同时间点比较,作用30 min时7 dynes/cm_(2)组的细胞面积显著大于12 dynes/cm_(2)组(P<0.05),而作用15、60和120 min时差异均无统计学意义(P>0.05)。结论:流体剪切应力为7 dynes/cm_(2)和12 dynes/cm_(2)作用下力学信号均能引起成骨细胞在形态学上发生效应性改变。 Objective:To observe the effects of fluid shear stress(FSS)with different strength on the morphology of osteoblasts.Methods:MG-63 osteoblasts cultured on the slides were exposed to shear stress under short exposures to fluid flow.HE staining and phase-contrast microscope were applied to observe the changes of the cell morphology and to analyze the influences of mechanical signal during the process under FSS of 7 dynes/cm_(2) and 12 dynes/cm_(2) in the time durations of 15,30,60 and 120 min,respectively.Results:When exposed to the 7 and 12 dynes/cm_(2) FSS respectively within 60 min,the morphology of the osteoblasts changed,with enhanced cell polarity and more regular arrangement.However,when exposed to FSS for 120 min,and the morphology of osteoblasts became irregular,with more abnormal mitosis.When exposed to 7 dynes/cm_(2) FSS for 30,60 and 120 min,and to 12 dynes/cm_(2) FSS for 60 and 120 min,the average cell area of osteoblasts increased(P<0.05).At 30 min time point,the average cell area of osteoblasts in 7 dynes/cm_(2) FSS group was significantly higher than that in 12 dynes/cm_(2) FSS group(P<0.05).Conclusion:The fluid shear stresses of 7 dynes/cm_(2) and 12 dynes/cm_(2) can influence the morphology of osteoblasts.
