大黄水煎液对家兔离体空肠平滑肌收缩功能的影响

Effect of Rhubarb Decoction on Contractile Function of Isolated Jejunal Smooth Muscle in Rabbits

目的基于大黄“泻下攻积”的功效,取实验用家兔离体空肠平滑肌,通过制备不同浓度的大黄水煎液,探讨其对平滑肌收缩作用的影响及可能的作用机制。方法取实验用家兔离体空肠平滑肌,采用平滑肌灌流的方法,观察在不同浓度条件下大黄水煎液对平滑肌收缩作用的影响。结果浓度为0.005 g/mL、0.01 g/mL大黄水煎液组与正常对照组比较,收缩幅度明显增加(P<0.05);浓度为0.005 g/mL、0.01 g/mL、0.015 g/mL大黄水煎液组与正常对照组比较,收缩频率明显加快(P<0.01);浓度为0.01 g/mL大黄水煎液可以改善阿托品对家兔离体空肠平滑肌收缩功能的抑制作用。结论不同浓度条件下的大黄水煎液,作用于家兔离体空肠平滑肌,均能明显促进平滑肌的收缩频率和收缩幅度,而且,随着浓度增加其促进作用先增强后减弱。同时,大黄水煎液可改善阿托品对家兔离体空肠平滑肌的抑制作用。

Objective Based on the effect of rhubarb to"reduce and attack the accumulation",to investigate the effects of different concentrations of rhubarb decoction on the contractile function of isolated jejunal smooth muscle in rabbits and its possible mechanism.Methods The effect of different concentrations of rhubarb decoction on the contractile function of rabbit isolated jejunum smooth muscle was observed and analyzed by perfusion method.Results Compared with normal control group,the systolic amplitude of rhubarb decoction groups with concentrations of 0.005 g/mL and 0.01 g/mL was significantly increased(P<0.05).The systolic frequency of rhubarb decoction group with concentrations of 0.005 g/mL,0.01 g/mL and 0.015 g/mL was significantly faster than that of normal control group(P<0.01).The concentration of 0.01 g/mL rhubarb decoction can improve the inhibitory effect of atropine on the contractile function of rabbit jejunal smooth muscle in vitro.Conclusions Different concentrations of rhubarb decoction can obviously promote the contractile frequency and amplitude of rabbit jejunal smooth muscle in vitro,and the promoting effect first increases and then decreases with the increase of concentration.At the same time,rhubarb decoction can improve the inhibitory effect of atropine on rabbit jejunal smooth muscle in vitro.
