摘要
以双水相法萃取分离红高粱色素,选取聚乙二醇(PEG)/(NH_(4))_(2)SO_(4)组成的双水相体系从高粱粒中萃取红高粱色素,确定双水相的组成成分PEG/(NH 4)2 SO 4对色素的影响。采用单一变量法,分别测定PEG分子量、PEG质量分数、(NH_(4))_(2)SO_(4)质量分数、水浴温度、粗提取液体积萃取红高粱色素的最佳值,按照方案进行响应面分析,得到萃取高粱色素的最佳条件:PEG2000质量为1.8 g,(NH_(4))_(2)SO_(4)质量为1 g,水浴温度为30℃,粗提取液体积为1.0 mL,加蒸馏水定量到10 g时,红高粱色素的萃取率最高。
Red sorghum pigment is extracted from sorghum grains with polyethylene glycol(PEG)/(NH_(4))_(2)SO_(4) aqueous two-phase system,and the effect of PEG/(NH_(4))_(2)SO_(4) on the pigment is determined.The molecular weight of PEG,mass fraction of PEG,mass fraction of (NH_(4))_(2)SO_(4),water bath temperature and crude extract volume are determined by single variable method to extract the best value of red sorghum pigment.Response surface analysis is carried out according to the scheme,and the optimal extraction conditions of sorghum pigment are obtained as follows:when the mass of PEG2000 is 1.8 g,the mass of (NH_(4))_(2)SO_(4) is 1 g,the water bath temperature is 30℃,and the crude extract volume is 1.0 mL,adding distilled water to 10 g,the extraction yield of red sorghum pigment is the highest.
