cGAS-STING信号通路及其触发因素与治疗药物 被引量:2

Trigger factors and therapeutic drugs of cGAS-STING signaling pathway

摘要 环状鸟苷单磷酸-腺苷单磷酸合酶(cyclic GMP-AMP synthase, cGAS)是一种能够与dsDNA结合的胞质核酸传感器。在无dsDNA情况下,cGAS保持抑制状态;在cGAS激活时,衔接蛋白干扰素基因刺激蛋白(stimulator of interferon genes, STING)被激活,触发级联反应,产生促炎细胞因子和Ⅰ型干扰素(typeⅠinterferon,Ⅰ-IFN)。cGAS-STING信号通路能够增强机体对感染的抵抗力,但是cGASSTING信号通路的过度激活可能会导致严重的炎症反应,影响自身免疫甚至产生疾病。本文系统回顾了cGAS-STING信号通路及其触发因素,并探讨了其与自身免疫性疾病的联系和药物治疗的进展。 Cyclic GMP-AMP synthase(cGAS) is a cytoplasmic nucleic acid sensor which can bind to dsDNA. cGAS remained inhibitory in the absence of dsDNA. But when cGAS is activated, stimulator of interferon gene(STING) is activated, triggering cascade reaction, producing proinflammatory cytokines and type Ⅰ interferon(Ⅰ-IFN). The cGAS-STING signaling pathway can enhance the body’s resistance to infection, but the excessive activation of cGAS-STING signaling pathway may lead to serious inflammation, affect autoimmune and produce diseases. This paper systematically reviewed the cGAS-STING signaling pathway and trigger factors, as well as its association with autoimmune diseases and drug treatment progress.
