摘要
为研究六羟基黄酮对金黄色葡萄球菌、奇异变形杆菌、大肠埃希氏菌及沙门氏菌的抑菌效果及其抗氧化性。本试验采用牛津杯法和微量肉汤稀释法,探讨六羟基黄酮对四种常见有害菌的抑菌能力;以维生素C(VC)为对照,应用水杨酸法和DPPH法,测定六羟基黄酮对羟基自由基(·OH)和DPPH的清除率,判断其抗氧化性能。结果表明:(1)六羟基黄酮对金黄色葡萄球菌、奇异变形杆菌、大肠埃希氏菌及沙门氏菌均具有较强的抑菌活性,浓度为100 mg/mL时抑菌效果最好;(2)六羟基黄酮对奇异变形杆菌的抑菌圈直径为23.30 mm,最低抑菌浓度为0.125 mg/mL;对大肠埃希氏菌抑菌圈直径为19.80 mm,最小抑菌浓度为0.625 mg/mL;对金黄色葡萄球菌抑菌圈直径为16.64 mm,最小抑菌浓度为0.25 mg/mL;对沙门氏菌抑菌圈直径为14.01 mm;最低抑菌浓度为2.5 mg/mL;(3)六羟基黄酮对·OH最高清除率为86.67%;对DPPH的最高清除率为96.69%。试验结果表明,六羟基黄酮对四种有害菌均有较强的抑制作用,且具有较好的抗氧化性,在饲料领域具有广阔的应用前景。
The purpose of this experiment was to study the antibacterial activity of 6-hydroxyflavone on Staphylococcus aureus,Proteus mirabilis,Escherichia coli,Salmonella and antioxidant activity.The antibacterial activity was studied by Oxford Cup method and micro-broth dilution method.In order to determine the antioxidant activity of 6-hydroxyflavone,the ascorbic acid(VC)as control,the salicylic acid method and DPPH method were used.The results showed that:(1)6-hydroxyflavone has strong antibacterial activity against Staphylococcus aureus,Proteus mirabilis,Escherichia coli and Salmonella,and the best concentration was 100 mg/mL.(2)The inhibition zone diameters of 6-hydroxyflavone against Proteus mirabilis,Escherichia coli,Staphylococcus aureus and Salmonellawere 23.30,19.80,16.64,14.01 mm respectively.The minimum inhibitory concentrations for the Proteus mirabilis,Escherichia coli,Staphylococcus aureus and Salmonella were 0.125,0.625,0.25,2.5 mg/mL respectively.(3)The highest clearance rate of 6-hydroxyflavone on hydroxyl and DPPH free radical was 86.67%and 96.69%respectively.The test showed that 6-hydroxyflavone has strong antibacterial and antioxidant activity and will be used broadly in husbandry.
