HPLC同步检测四味穿心莲散中绿原酸和芦丁的含量

Simultaneous determination of chlorogenic acid and rutin in compound Chinese medicine Siweichuanxinlian powder by HPLC

为建立HPLC方法同步测定四味穿心莲散中绿原酸和芦丁的方法,本实验以甲醇为提取溶剂,乙腈-0.1%磷酸水为流动相进行梯度洗脱,C18色谱柱分离,355 nm波长检测。结果显示:绿原酸、芦丁浓度在0.1~0.9 mg/L内呈良好的线性关系,加样回收率分别为98.41%、98.12%;重复性良好,相对标准偏差<1%。可见本方法适用于四味穿心莲复方中绿原酸和芦丁含量测定。

A high performance liquid chromatography(HPLC)method was developed for the simultaneous quantitation of chlorogenic acid and rutin in compound Chinese medicine Siweichuanxinlian.The samples were extracted in methanol,the HPLC separation was achieved on a C18 column using a mobile phase of acetonitrile:0.1%phosphate acid water.The data collection was set at 355 nm.The method provided good linearity from 0.1~0.9 mg/L.The mean recoveries(n=6)of chlorogenic acid and rutin in samples were 98.41%and 98.12%,respectively and there was a good repeatability(RSD<1%).It was a simple,rapid and reliable method for the simultaneous determination of chlorogenic acid and rutin in compound Chinese medicine Siweichuanxinlian.
