青钱柳黄酮对幼年免疫低下并肥胖大鼠血糖、血脂、免疫功能的调节作用 被引量:5

摘要 目的探讨青钱柳黄酮对幼年免疫低下并肥胖大鼠血糖、血脂、免疫功能的调节作用。方法将2周龄SD大鼠分为对照组、高脂组、环磷酰胺+高脂组、治疗组Ⅰ、治疗组Ⅱ、治疗组Ⅲ,除对照组给予正常饮食外其他组均给予高脂饲料喂养。同时,环磷酰胺+高脂组给予环磷酰胺灌胃,3个治疗组给予环磷酰胺和相应剂量青钱柳黄酮(20 mg/kg、40 mg/kg、80 mg/kg)灌胃。干预15 d后,记录大鼠体重,并检测血脂、血糖、胸腺指数、肝脏指数和脾脏指数、脾淋巴细胞增殖率以及腹腔白色脂肪组织中磷酸化核因子κB p65(p-NF-κB p65)的表达情况。结果(1)与对照组比较,高脂组和环磷酰胺+高脂组的大鼠终体重均增加,血糖、总胆固醇、三酰甘油、LDL-C水平、肝脏指数、脂肪组织内p-NF-κB p65表达水平均升高,血清HDL-C水平均降低(P<0.05);高脂组、对照组、环磷酰胺+高脂组的脾淋巴细胞增殖率、胸腺指数及脾脏指数依次降低(均P<0.05)。(2)环磷酰胺+高脂组、治疗组Ⅰ、治疗组Ⅱ、治疗组Ⅲ的血清总胆固醇、三酰甘油、LDL-C水平、血糖水平、肝脏指数依次降低,脾淋巴细胞增殖率、胸腺指数、脾脏指数均依次升高(均P<0.05)。与环磷酰胺+高脂组比较,3个治疗组的p-NF-κB p65表达水平均下调(均P<0.05)。结论青钱柳黄酮可剂量依赖性地改善幼年免疫低下并肥胖大鼠的血糖、血脂和免疫异常。 Objective To explore the regulation effects of Cyclocarya paliurus flavonoids on blood glucose,blood lipid,and immune function in infant rats with obesity and immunosuppression.Methods Two-week-old SD rats were divided into control group,high-fat group,cyclophosphamide(CTX)+high-fat group,treatment groupⅠ,treatment groupⅡand treatment groupⅢ,and except the control group with normal diet,the remaining groups were fed by high-fat diet.Meanwhile,the CTX+high-fat group was intragastrically administrated CTX,and the three treatment groups were intragastrically administrated CTX and Cyclocarya paliurus flavonoids of corresponding concentrations(20 mg/kg,40 mg/kg,and 80 mg/kg).After 15 days of intervention,the weight was recorded in all rats,besides,blood lipid,blood glucose,thymus index,liver index,spleen index,spleen lymphocyte proliferation rate,and the expression of phosphorylated nuclear factorκB p65(p-NF-κB p65)in abdominal white adipose tissues was determined.Results(1)Compared with the control group,the high-fat group and the CTX+high-fat group yielded an increased final weight,higher blood glucose,TC,TG and LDL-C levels,elevated liver index and p-NF-κB p65 expression level in adipose tissues,and lower serum HDL-C level(all P<0.05);the spleen lymphocyte proliferation rate,thymus index and spleen index decreased in the sequence of the high-fat group,the control group,and the CTX+high-fat group(all P<0.05).(2)Serum TC,TG and LDL-C levels,blood glucose level and liver index decreased,and the spleen lymphocyte proliferation rate,thymus index and spleen index increased in the CTX+high-fat group,the treatment groupⅠ,the treatment groupⅡand the treatment groupⅢsuccessively(all P<0.05).Compared with the CTX+high-fat group,the expression level of p-NF-κB p65 was down-regulated in the three treatment groups(all P<0.05).Conclusion Cyclocarya paliurus flavonoids can improve blood glucose,blood lipid and immune abnormality in a dose-independent manner in infant rats with obesity and immunosuppression.
