外源NO和ABA处理对马铃薯块茎创伤周皮中软木脂积累的影响 被引量:1

摘要 【目的】愈伤是马铃薯块茎受损后的典型反应,对块茎采后减轻病害有重要作用。但块茎完全愈伤所需的时间较长,为寻求促进愈伤的措施,实验研究外源NO和ABA处理对马铃薯块茎创伤周皮中软木脂积累的影响,并探索薯块创伤后的愈合机制。【方法】以马铃薯品种"网纹108"和"滇薯47"为材料,进行去周皮和不去周皮两等份处理,每个马铃薯用刀切成8小块,以切面为创伤面。实验设对照(超纯水);处理1(1 mmol·L^(-1 )SNP);处理2(2 mmol·L^(-1) SNP);处理3(0.13 mmol·L^(-1) cPTIO);处理4(0.1 mmol·L^(-1) ABA)5个处理。马铃薯块茎创伤处理后,置于室温(22±1℃,湿度85%)进行创伤愈合反应。于创伤后24、72、120和168 h取样染色观察。【结果】块茎愈伤期间,软木脂的积累量随愈伤时间的延长而不断增加,SNP处理和ABA处理的软木脂积累速率显著高于对照,对2个品种马铃薯均有显著促进软木脂积累的作用(P<0.05)。其中,SNP处理中以2 mmol·L^(-1)效果更佳,其处理的薯块软木脂累积在168 h时较其他处理的高,而ABA在72 h时响应较好。其次,不同品种创伤后软木脂的累积存在显著差异,"W108"软木脂的累积显著高于"D47"。另外,去除原周皮对块茎软木脂的累积存在一定的影响,"D47"创伤时保留原周皮的块茎软木脂累积更多,而"W108"去除原周皮的块茎软木脂累积更多。【结论】外源NO和ABA处理能显著促进马铃薯创伤后软木脂的积累,表明外源NO和ABA处理能促进块茎愈伤。不同品种软木脂的积累存在差异,原周皮对创伤后软木脂的累积因品种不同存在差异。 【Objective】Healing is a typical response of potato tubers after wounding,which plays an important role in reducing postharvest diseases of tubers.However,it takes a long time for the tubers to completely heal.In order to seek measures to promote the healing,the effect of exogenous NO and ABA treatments on the accumulation of suberin in the wound-healing periderm of tubers was studied experimentally,and the wound-healing mechanism of potato tubers after wounds was explored.【Method】In this study,potato tubers from‘W108’and‘D47’were used as materials.The materials were divided into two parts,peeled and unpeeled.Each potato was cut into 8 small pieces,using the cut surface as the wound surface.The artificially damaged tubers were treated with five treatment:Control(ultra-pure water);Treatment 1(1 mmol·L^(-1) SNP);Treatment 2(2 mmol·L^(-1) SNP);Treatment 3(0.13 mmol·L^(-1) cPTIO);Treatment 4(0.1 mmol·L^(-1) ABA).The accumulation of suberin was observed at the wound site during wound-healing.【Result】The accumulation of suberin gradually increased in wounded tubers during healing.SNP treatment and ABA treatment significantly promoted the accumulation of suberin in both varieties of potato(P<0.05)than the control.Treatment 2(2 mmol·L^(-1) SNP)had a better effect,which the accumulation of suberin was higher at 168 hours than in other treatments,while ABA had a better response at 72 hours.Then,there was a significant difference in the accumulation of suberin among different varieties,and the accumulation of suberin in‘W108’was significantly higher than that in‘D47’during wound-healing.In addition,the removal of native periderm had a certain effect on the accumulation of suberin in tubers.During wound-healing,the unpeeled tubers of‘D47’accumulated more suberin,while the peeled tubers of‘W108’accumulated more suberin.【Conclusion】Exogenous NO and ABA treatments could significantly promote the accumulation of suberin after wound,that indicated exogenous NO and ABA treatments promotes wound healing of potato tubers.There were differences in the accumulation of suberin among different potato varieties and the native periderm.
