促生菌接种对青稞根际土壤微生物群落结构的影响 被引量:7

摘要 【目的】本研究探讨了促生菌浸种处理对青稞根际土壤微生物群落结构和菌群多样性的影响,旨在为促生菌在青稞生产中应用提供理论依据。【方法】利用前期筛选出的2株促生菌TSXJ4与GSXJ进行浸种处理,于青稞灌浆期采集根际土壤样品,采用MiSeq高通量测序技术,分析鉴定青稞根际土壤微生物群落结构和菌群多样性。【结果】促生菌浸种改变了青稞根际土壤中细菌及真菌的群落结构,真菌群落结构的变化较为明显。根际土壤细菌丰富度和多样性表现为GSXJ4>TSXJ4>CK,真菌丰富度和多样性结果与细菌相反;细菌群落中影响较大的为Actinobacteria及Sphingobacteriia,通过浸种处理,Actinobacteria数量明显上升,而Sphingobacteriia数量明显下降;在真菌群落中影响较大的为Agaricomycetes及Pezizomycetes,通过浸种处理,Agaricomycetes数量明显下降,而Pezizomycetes,数量明显上升。【结论】促生菌浸种提高了青稞根际土壤中细菌的丰富度和多样性,而降低了真菌的丰富度和多样性;青稞根际土壤放线菌纲微生物反应对促生菌浸种较为敏感,且真菌各菌群数目变化波动较大;促生菌浸种使青稞根际土壤细菌种群多样性增加,但优势菌群种类及数量较稳定,且真菌种类变化较细菌明显。 【Objective】The present paper aimed to study the effect of seed soaking treatment on the hulless barley rhizosphere soil microbial community structure and flora diversity,which would provide a theoretical basis for the application.【Method】TSXJ4 and GSXJ,two strains of growth promoting bacteria screened in the previous stage,were used for seed soaking treatment,rhizosphere soil samples were collected at the hulless barley grouting stage,and MiSeq high-throughput sequencing technology was adopted to analyze and identify the microbial community structure and microbial diversity of the hulless barley rhizosphere soil.【Result】The growth promoting bacteria immersion species changed the community structure of bacteria and fungi in the hulless barley rhizosphere soil,and the change of fungal community structure was relatively obvious.The bacteria richness and diversity of rhizosphere soil were GSXJ4>TSXJ4>CK,and the results of fungal richness and diversity were contrary to that of bacteria.Actinobacteria and Sphingobacteriia had a great effect on the bacterial community,the number of Actinobacteria increased significantly,and the number of Sphingobacteriia decreased significantly after soaking in the species.Agaricomycetes and Pezizomycetes had a greater effect on the fungal community.Agaricomycetes decreased,and Pezizomycetes increased after species infusion.【Conclusion】The growth promoting bacteria immersion increased the bacterial richness and diversity in the huless barley rhizosphere soil,but decreased the fungal richness and diversity.The microbial reactions of the soil actinomycetes in the hulless barley rhizosphere were sensitive to the growth promoting bacteria,and the number of fungi fluctuated greatly.The species diversity of the hulless barley rhizosphere soil bacteria was increased by seeding with probiotic bacteria,but the species and number of dominant bacteria were more stable,and the change of fungal species was more obvious than that of bacteria.
