Effect of Different Cryoprotectants on Spermatozoa Cryopreservation of Siberian Sturgeon(Acipenser baerii baeri)

摘要 【目的】研究不同抗冻剂对西伯利亚鲟精子的冷冻保存效果,筛选出合适的鲟鱼精子超低温冷冻保存抗冻剂。【方法】采用计算机辅助精子分析系统(CASA),检测10%、15%、20%的甲醇(MeOH)和二甲基亚砜(DMSO)分别作为抗冻剂超低温保存西伯利亚鲟精液活力及运动参数差异。【结果】以MeOH和DMSO作为抗冻剂,在浓度分别为10%、15%、20%条件下,精子解冻激活后DMSO保存的精子活力及精子的运动参数均高于MeOH保存的精子。各处理抗冻效果以15%DMSO最优,其A+B级精子活力占比最高,为44.83%;精子的平均曲线运动速度(VCL)、平均路径速度(VAP)最高,分别为24.33、22.16μm/s,显著高于10%DMSO组精子,与20%DMSO间差异不显著;精子头侧摆幅度(ALH)、运动的摆动性(WOB)、平均移动角度(MAD)指标3种浓度间差异不显著。【结论】西伯利亚鲟的精子超低温冷冻保存抗冻剂以15%DMSO的抗冻效果最好。 【Objective】The present study was to research the effects of different concentrations of cryoprotectants on the sperm cryopreservation of Siberian sturgeon(Acipenser baerii baeri),and to screen suitable sperm cryoprotectants.【Method】The CASA(Computer Assisted Semen Analysis)was employed to determine the differences of sperm motility and kinematic parameters using three concentrations(10%,15%,20%)of methanol and DMSO as cryoprotectants.【Result】With MeOH and DMSO as antifreeze agents,the motility and motility parameters of sperm preserved by DMSO after thawing and activation were higher than those preserved by MeOH at concentrations of 10%,15%and 20%,respectively.The antifreeze effect of all treatments was the best with 15%DMSO,and the proportion of A+B sperm motility was the highest,44.83%.The mean curve velocity(VCL)and mean path velocity(VAP)of sperm were the highest,which were 24.33 and 22.16μm/s,respectively,which were significantly higher than those of 10%DMSO group,but had no significant difference from 20%DMSO group.There were no significant differences among ALH,WOB and MAD concentrations of sperm.【Conclusion】15%DMSO was the best cryoprotectants of Siberian sturgeon.
