非小细胞肺癌中RABEP1的表达及与临床病理特征和预后的关系 被引量:3

Expressions of RABEP1 in non-small cell lung cancer and their relationships with clinicopathological features and prognosis

摘要 目的通过检测Rab GTP结合效应蛋白(RABEP1)在非小细胞肺癌中表达变化,探究其与非小细胞肺癌患者临床病理特征及预后的关系。方法前瞻性选择2015年2月至2018年2月在琼海市人民医院胸外科治疗的非小细胞肺癌患者82例,术中收集非小细胞肺癌组织及相应癌旁组织(距肿瘤组织>3 cm)。在Ualcan数据库中检索RABEP1表达情况。采用免疫组织化学染色法检测组织中RABEP1表达水平,分析其与非小细胞肺癌临床特征关系。对入组患者进行3年随访,采用Kaplan-Meier法分析患者生存情况。采用Cox回归模型分析RABEP1蛋白表达与非小细胞肺癌患者预后关系。结果Ualcan数据库中,RABEP1在肺鳞癌和肺腺癌组织中的表达分别为(16.23±5.40)、(14.99±4.95),均明显低于正常组织[(19.65±6.49)、(19.44±6.46)],差异均有统计学意义(P<0.05)。非小细胞肺癌组织中RABEP1蛋白高表达率(35.37%)明显低于癌旁组织(60.98%),差异有统计学意义(P<0.05)。RABEP1表达与肿瘤大小、分化程度、有无浸润肺膜、TNM分期、是否有淋巴结转移有关(P<0.05)。本次随访患者生存33例,死亡45例,失访4例,随访率95.12%。RABEP1低表达组患者生存16例,RABEP1高表达组患者生存19例。RABEP1低表达组生存率(34.48%)显著低于RABEP1高表达组(69.81%),差异有统计学意义(P<0.05)。浸润肺膜(95%CI:3.612~21.034)、TNM分期为Ⅲ期(95%CI:1.364~6.053)、淋巴结转移(95%CI:1.891~8.572)、RABEP1低表达(95%CI:1.563~9.090)是影响非小细胞肺癌患者不良预后的独立危险因素(P<0.05)。结论非小细胞肺癌组织中RABEP1低表达,与肿瘤临床分期、淋巴结转移等临床病理特征及生存率有关,可能影响非小细胞肺癌进展及患者预后。 Objective To investigate the expression of Rab GTPase-binding effector protein(RABEP1)protein in non-small cell lung cancer and its relationship with clinicopathological features and prognosis.Methods A tatol of 82 patients with non-small cell lung cancer who were treated in Department of Thoracic Surgery,Qionghai People's Hospital from February 2015 to February 2018 were prospectively selected.Non-small cell lung cancer tissues and corresponding adjacent tissues(more than 3 cm away from the tumor)were collected during operation.The expression of RABEP1 was searched in Ualcan database.The expression levels of RABEP1 in tissues were detected by immunohistochemical staining,and their relationships with clinical characteristics of non-small cell lung cancer were analyzed.Cox regression model was used to analyze the relationships between the expressions of RABEP1 and the prognosis of NSCLC patients.Results In the Ualcan database,the expression of RABEP1 in lung squamous cell carcinoma and lung adenocarcinoma tissues were(16.23±5.40)and(14.99±4.95),which were significantly lower than those in normal tissues[(19.65±6.49),(19.44±6.46)],the differences were statistically significant(P<0.05).The high expression rate of RABEP1 protein in NSCLC tissues(35.37%),which was significantly lower than that in adjacent tissues(60.98%),the differences were statistically significant(P<0.05).RABEP1 expression was correlated with tumor size,differentiation degree,infiltration of pulmonary membranes,TNM stage and lymph node metastasis,the difference was statistically significant(P<0.05).In this follow-up,33 patients survived,45 died and 4 were lost to follow-up,with a follow-up rate of 95.12%.There were 16 patients in the RABEP1 low expression group survived but 19 in the RABEP1 high expression group.The survival rate of RABEP1 low expression group(34.48%)was significantly lower than that of RABEP1 high expression group(69.81%),the difference was statistically significant(P<0.05).Infiltration of pulmonary membranes(95%CI:3.612~21.034),TNM stage was stage III(95%CI:1.364~6.053),lymph node metastasis(95%CI:1.891~8.572),low expression of RABEP1(95%CI:1.563~9.090)were independent risk factors for poor prognosis of non-small cell lung cancer(P<0.05).Conclusion The expression of RABEP1 is low and the expression of in non-small cell lung cancer,which is related to clinical pathological features and survival rate,such as clinical stage,lymph node metastasis,and may affect the progress and prognosis of non-small cell lung cancer.
