樱桃查尔酮合成酶CpCHS2基因的克隆及序列分析 被引量:1

摘要 查尔酮合成酶基因是类黄酮生物合成的关键基因,本文通过巢氏PCR,从‘玛瑙红’樱桃的根系组织中克隆获得一个1176 bp的查尔酮合成酶基因的cDNA序列,编码391个氨基酸,命名为CpCHS2(基因登录号:MT112906)。该基因编码的蛋白分子量为42.68 kD,等电点为5.76,属疏水性蛋白且不具有跨膜区,主要定位在细胞质中,二级结构中α-螺旋、β-折叠为蛋白质最大量的结构原件。系统进化树分析表明,CpCHS2和同属蔷薇科的红叶李及碧桃的亲缘关系最近,并在进化上高度保守。 Chalcone synthetase gene is the key gene for flavonoid biosynthesis.In this paper,a full-length cDNA of the CHS gene was obtained by Nested PCR from the root tissue of Cerasus pseudocerasus‘Manaohong’,whose complete cDNA was 1,176 bp,encoding 319 amino acids,and being designated as CpCHS2(Gene accession number is MT112906).The protein encoded by the gene has a molecular weight of 42.68 kD and an isoelectric point of 5.76.It was a hydrophobic protein and did not have a transmembrane region,which was mainly located in the cytoplasm.In the secondary structure,α-helix andβ-sheet were the largest structural elements of the protein.The phylogenetic tree analysis showed that the CpCHS2 and Prunus cerasifera and Amygdalus persica of the same genus Rosaceae were closely related and highly conserved in evolution.
