摘要
目的探讨miR-93是否通过调节Bax/Bcl-2的表达活性对胃癌细胞增殖、迁移和凋亡产生影响及其作用机制。方法qPCR法检测人正常胃细胞和胃癌细胞中miR-93的表达情况;克隆形成实验检测miR-93对胃癌细胞增殖能力的影响;细胞划痕实验检测miR-93对胃癌细胞迁移能力的影响;流式细胞术检测miR-93对胃癌细胞凋亡能力的影响;qPCR和Western blot法检测miR-93对胃癌细胞中Bax和Bcl-2 mRNA和蛋白表达的影响;流式细胞仪检测miR-93对胃癌细胞中Caspase-3活性的影响。结果与人正常胃细胞组相比,胃癌细胞中miR-93表达水平显著上调(P<0.01);与control组和miR-93-NC组相比,miR-93-inhibitor组SGC-7901细胞增殖能力、迁移能力均下降(P<0.01,P<0.05),凋亡能力增强(P<0.01);与control组和miR-93-NC组相比,miR-93-inhibitor组SGC-7901细胞中Bax mRNA和蛋白的表达上调(P<0.01),Bcl-2 mRNA和蛋白的表达下调,且Bax/Bcl-2的比例上调(P<0.001),Caspase-3的活性提高。结论下调miR-93调节Bax/Bcl-2的表达活性对胃癌细胞增殖、迁移及凋亡的影响。
Objective To investigate to investigate whether miR-93 can affect the proliferation,migration and apoptosis of gastric cancer cells by regulating Bax/Bcl-2 expression and its mechanism.Methods qPCR was used to detect the expression of miR-93 in human normal gastric and gastric cancer cells;The colony formation assay was used to detect the effect of miR-93 on the proliferation of gastric cancer cells;Cell scratch test was used to detect the effect of miR-93 on the migration ability of gastric cancer cells;Flow cytometry was used to detect the effect of miR-93 on the apoptosis ability of gastric cancer cells;The effects of miR-93 on the expression of Bax and Bcl-2 mRNA and protein in gastric cancer cells were detected by qPCR and Western blot;Flow cytometry was used to detect the effect of miR-93 on Caspase-3 activity in gastric cancer cells.Results The expression level of miR-93 was significantly up-regulated in gastric cancer cells compared with human normal gastric cells(P<0.01);Compared with the control group and the miR-93-NC group,the proliferation and migration ability of SGC-7901 cells was decreased in the miR-93-inhibitor group(P<0.01,P<0.05),and the apoptosis ability increased(P<0.01);Compared with the control group and the miR-93-NC group,the expression of Bax mRNA and protein was up-regulated in the SGC-7901 cells of the miR-93-inhibitor group(P<0.01),and the expression of Bcl-2 mRNA and protein was down-regulated,and the proportion of Bax/Bcl-2 was up-regulated(P<0.001),and the activity of Caspase-3 was increased.Conclusion Down-regulating the expression of Bax/Bcl-2 by miR-93 can affect the proliferation,migration and apoptosis of gastric cancer cells.
