CYP27A1抑制膀胱癌细胞增殖及作用机制研究

摘要 目的研究CYP27A1对膀胱癌细胞的影响及其作用机制。方法 qRY-PCR检测膀胱癌细胞中AR和CYP27A1的mRNA水平;采用转染的实验技术使T24细胞稳定表达CYP27A1;MTT实验检测CYP27A1和27-HC对膀胱癌细胞增殖的影响;裸鼠移植瘤模型检测CYP27A1对膀胱癌细胞增殖的影响;ELIAE检测细胞上清液中27-HC的含量。结果 T24细胞中AR处于高表达水平CYP27A1处于低表达水平,5637细胞中AR处于低表达水平,CYP27A1处于高表达水平;在mRNA水平和蛋白质水平检测到CYP27A1在T24细胞中稳定表达;CYP27A1高表达的T24细胞的增殖能力明显减弱;CYP27A1高表达的T24细胞的上清液中27-HC的含量明显增加;27-HC可显著抑制膀胱癌细胞的增殖。结论 CYP27A1通过分泌27-HC抑制膀胱癌细胞的增殖。 Objective To study the effect of CYP27 A1 on bladder cancer cells and its mechanism.Methods qRYPCR was used to detect the mRNA levels of AR and CYP27 A1 in bladder cancer cells.The expression of CYP27 A1 was stably expressed in T24 cells by transfection technique.The effect of CYP27 A1 and 27-HC on the proliferation of bladder cancer cells was detected by MTT assay.The effect of CYP27 A1 on the proliferation of bladder cancer cells was examined;ELIAE was used to detect the content of 27-HC in the supernatant of cells.Results In the T24 cells,AR was at a high expression level,CYP27 A1 was at a low expression level,and in 5637 cells,AR was at a low expression level,CYP27 A1 was at a high expression level;CYP27 A1 was stably expressed in T24 cells at mRNA and protein levels;CYP27 A1 was highly expressed in T24.The proliferative capacity of the cells was significantly attenuated;the content of 27-HC in the supernatant of T24 cells with high expression of CYP27 A1 was significantly increased;27-HC and significantly inhibited the proliferation of bladder cancer cells.Conclusion CYP27 A1 inhibits the proliferation of bladder cancer cells by secreting 27-HC.
