红景天甙调控sirt1/FOXO1通路改善酒精性肝损伤的实验研究 被引量:1

摘要 目的探讨红景天甙对沉默信息调节因子1(sirt1)/叉头转录因子(FOXO1)通路的调控作用,及对酒精性肝损伤(ALI)大鼠肝损伤的保护作用。方法取SD大鼠建立ALI模型,并将其分为正常对照组、模型组、红景天甙组、sirt1抑制剂(Tenovin-6)组、红景天甙+Tenovin-6组,每组10只。检测肝组织损伤、肝功能指标、肝组织氧化应激指标,肝组织sirt1、FOXO1、乙酰化FOXO1(acFOXO1)、脂联素受体2(AdipoR2)、微粒体甘油三酯转运蛋白(MTP)蛋白表达水平。结果与模型组相比,Tenovin-6组大鼠肝组织损伤加重,肝功能指标、肝组织氧化应激水平、肝组织acFOXO1表达等进一步升高,sirt1、AdipoR2及MTP表达进一步降低(P<0.05);红景天甙组大鼠肝组织损伤最轻,上述指标变化与模型组相反(P<0.05)。红景天甙+Tenovin-6组上述指标变化与红景天甙组相反(P<0.05)。结论红景天甙可通过激活sirt1/FOXO1/AdipoR2/MTP通路,改善肝脏脂质代谢,减轻脂质堆积,保护ALI大鼠肝功能。 Objective To explore the regulation of salidroside on the silent information regulator 1(sirt1)/forkhead transcription factor(FOXO1)pathway,and its protective effect on alcoholic liver injury(ALI)rats.Methods Take SD rats to establish ALI model,and divide them into normal control group,model group,salidroside group,sirt1 inhibitor(Tenovin-6)group,salidroside+ Tenovin-6 group.There were 10 animals in each group.The liver tissue damage was detected;liver function index,liver tissue oxidative;liver tissue sirt1,FOXO1,acetylated FOXO1(acFOXO1),adiponectin receptor 2(AdipoR2),microsomal triglyceride transfer protein(MTP)protein The expression level.Results Compared with the model group,the liver tissue damage in the Tenovin-6 group was aggravated,liver function indexes,liver tissue oxidative stress level,liver tissue acFOXO1 expression,etc.were further increased,sirt1,AdipoR2 and MTP expression were further decreased,and each index changed(P<0.05);the liver tissue damage of rats in the salidroside group was the least,the changes in the above indicators were opposite to the model group(P<0.05).The changes of the above indexes in the salidroside+Tenovin-6 group were opposite to those in the salidroside group(P<0.05).Conclusion Salidroside can improve liver lipid metabolism,reduce lipid accumulation and protect liver function in ALI rats by activating sirt1/FOXO1/AdipoR2/MTP pathway.
