溴硝醇有关物质的测定

摘要 建立了测定溴硝醇有关物质的高效液相色谱方法,色谱柱为RP ODS(150 mm×4.6 mm,5μm)柱,其流动相为V(磷酸水)∶V(甲醇)=95∶5(磷酸水中,磷酸与水的体积比1∶1000),流速为1.0 mL/min,柱温为30℃,检测波长为210 nm,对溴硝醇开展了专属性、检测限、线性、精密度、再现性、稳定性和耐用性研究。结果表明:溴硝醇与其杂质良好分离,溴硝醇在质量浓度为2-40μg/mL区间内线性关系良好(相关系数R^(2)=0.9999),其平均回收率为102.46%(RSD=1.79%)。在本测定条件下溴硝醇的检测限为0.1μg/mL;定量限为1μg/mL。该方法具有操作简便、准确性高及专属性良好的特点,可用于溴硝醇有关物质的测定。 We established a high-performance liquid chromatography(HPLC)method to determine bronopol-related substances.The determination was performed on Agilent HPLC with RP-ODS(150 mm×4.6 mm,5μm)with the mobile phase of phosphoric acid-methyl alcohol(V∶V=95∶5)at a flow rate of 1.0 mL/min,a column temperature of 30℃,and a detection wavelength of 210 nm.In addition the phosphoric acid was composed of phosphoric acid and water at a ratio of 1∶1000(V∶V).The specificity,detection limit,linearity,precision,reproducibility,stability,and durability studies have been carried out for bronopol.The results show that bronopol was well separated from its impurity.Bronopol showed a great linear relationship at concentrations ranging from 2 to 40μg/mL(R^(2) was 0.9999).And the average recovery was 102.46%(RSD=1.79%).The detection limit of bronopol was 0.1μg/mL under this condition.Moreover the limit of quantitation was 1μg/mL.Our finding shows that the method is convenient,accurate and specific to determine bronopol-related substances.
