改良硅珠法检出19年血刀上陈旧血痕DNA分析1例 被引量:1

摘要 介绍1例通过改良硅珠法检出19年血刀上陈旧血痕DNA的方法。改良硅珠法采用《D盾超敏DNA提取试剂盒》,通过改良裂解液配方,加入EDTA(0.5mol/L)螯合血刀铁锈中的金属离子,在裂解30min后,通过更新裂解液以减少DNA氧化损伤,然后用硅珠法纯化提取DNA,经扩增和电泳检测,与试剂盒标准方法对比试验结果。试剂盒标准方法未检出DNA,改良硅珠法可得到完整的STR分型图谱,图谱的PCR产物峰高呈现小片段高、大片段低的趋势。本案例介绍的方法,对于陈旧、微量铁锈类检材DNA检验具有借鉴意义。
