摘要
对鹿筋进行深入加工、开发、利用,采用响应面法优化鹿筋胶原蛋白酶解工艺。以酶解温度、酶解时间、pH为自变量,鹿筋胶原蛋白水解度为因变量,进行单因素试验,采用Box-Behnken法进行响应面试验设计,确定鹿筋胶原蛋白的最佳酶解工艺。通过氨基酸分析,初步研究鹿筋胶原蛋白的特征性质。结果表明,最佳酶解工艺为pH 9.4,酶解时间5 h,酶解温度53℃(初始底物浓度3%、酶底比1%)。在此条件下,水解度为31.07%。氨基酸分析结果表明,鹿筋胶原蛋白的氨基酸组成中,甘氨酸含量最高,约占氨基酸总含量的30.25%,同时脯氨酸和羟脯氨酸含量也较高,分别占氨基酸总含量的8.24%和6.05%,符合胶原蛋白特征性质。
In order to further process,develop and utilize deer sinew,response surface methodology was used to optimize the enzymolysis process of deer sinew collagen.The enzymatic temperature,enzymatic time and pH were used as independent variables,and the degree of hydrolysis of deer sinew collagen was used as dependent variables.The Box-Behnken experiment design was used to obtain the optimum enzymolysis process of deer sinew.The characteristics of deer sinew collagen were studied by amino acid analysis.The optimum parameters of deer sinew collagen were pH 9.4,time 5 h,temperature 53℃(substrate concentration 3%,enzyme substrate ratio 1%),and the degree of hydrolysis was 31.07%.The results of amino acid analysis showed that the content of glycine was the highest,accounting for about 30.25%of the total amino acid content,while the content of proline and hydroxyproline were also higher,accounting for 8.24%and 6.05% of the total amino acid content,respectively,which was conformed to the characteristics of collagen.
