小鼠心脏发育中RNA编辑的动态变化及影响

摘要 目的探讨RNA编辑在小鼠心脏发育过程中的动态变化并对其功能影响进行分析预测。方法从ArrayExpress数据库获取小鼠心脏发育的转录组数据(E-MTAB-6798),使用SPRINT和ANNOVAR工具识别和注释RNA编辑位点,从编辑酶表达水平、编辑位点数、样本编辑水平和编辑指数方面描述RNA编辑的动态变化,并通过差异编辑及共编辑网络分析探究RNA编辑的功能和意义。结果在55个样本中共识别出122554个非冗余的RNA编辑位点,样本的编辑水平和编辑指数在小鼠心脏发育过程中变化明显,且出生两周后样本中的RNA编辑更加特异。心脏发育过程中差异编辑的位点可能通过影响miRNA与mRNA的结合调控着免疫相关基因的表达。与心脏整个发育过程显著相关的共编辑模块显示与趋化因子配体2的产生和编码心脏肌球蛋白α重链亚基的Myh6基因有关。结论RNA编辑可能影响着小鼠心脏发育过程中的免疫活动,并且影响着Myh6等基因的正确编码。 ObjectiveTo explore the dynamic changes of RNA editing during mouse heart development and to predict its functional effects.MethodsThe transcriptome sequencing data of mouse heart development was collected from ArrayExpress(E-MTAB-6798),and the RNA editing sites were identified by SPRINT and annotated by ANNOVAR. The dynamic changes of RNA editing were analyzed according to the expression level of editing enzymes,the number of editing sites,and the editing level and the editing index of samples. And through differential editing and co-editing network analysis,the functions of RNA editing during mouse heart development were predicted.Results editing sites were identified from 55 RNA-seq data. The editing level and editing index of the samples changed significantly during heart development,and the RNA editing in samples two weeks post-natal was more developmental specific. Differentially edited sites during heart development might regulate the expression of immune-related genes by influencing the binding of miRNA and mRNA. Two co-edited modules,which were significantly related to the entire development process of mouse heart,showed that they were related to the production of chemokine ligand 2 and the Myh6 gene encoding the cardiac myosin alpha heavy chain subunit.ConclusionRNA editing might affect the immune activity during mouse heart development,and might affect the correct coding of genes like Myh6.
