益智解毒汤对糖尿病大鼠认知功能障碍及糖脂代谢的影响 被引量:1

摘要 目的探讨益智解毒汤对糖尿病大鼠学习记忆能力、糖脂代谢及胰岛素分泌的影响。方法将48只SD大鼠随机分为正常对照组、糖尿病模型组、益智解毒汤低剂量组、益智解毒汤中剂量组、益智解毒汤高剂量组和盐酸多奈哌齐组,每组8只。除正常对照组外,其余5组均给予高糖高脂饮食联合链脲佐菌素诱发糖尿病。益智解毒汤低、中、高剂量组分别给予益智解毒汤0.9、1.8、3.6 g/(kg·d)灌胃,盐酸多奈哌齐组予盐酸多奈哌齐0.45 mg/(kg·d)灌胃,正常对照组及糖尿病模型组给予等量生理盐水。连续灌胃4周后,测定大鼠体质量、空腹胰岛素(FINS)、空腹血糖(FBG)、血清总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、甘油三酯(TG),并观察大鼠行为学改变及海马区神经元的病理变化。结果与糖尿病模型组相比,益智解毒汤高剂量组大鼠体质量得以控制(P<0.05),FBG、TG、LDL-C明显降低(P<0.01,P<0.05);大鼠逃避潜伏期显著缩短(P<0.05),跨越平台次数增加(P<0.05);病理可见细胞核固缩减少,变性坏死细胞数量减少,尼氏小体数量增加。结论益智解毒汤可显著改善糖尿病大鼠的糖脂代谢,促进胰岛素分泌,改善认知功能障碍。 Objective To explore the effect of Yizhi Jiedu Decoction on the learning and memory ability,glucose and lipid metabolism and insulin secretion in diabetic rats.Methods Forty-eight SD rats were randomly divided into normal control group,diabetes model group,low-dose,medium-dose,high-dose Yizhi Jiedu Decoction groups,and donepezil hydrochloride groups,each with eight rats.Except for the normal control group,the other five groups were given a high-sugar and high-fat diet combined with streptothricin to induce diabetes.low-dose,medium-dose,high-dose Yizhi Jiedu Decoction groups were given orally by 0.9,1.8,3.6 g/(kg·d)respectively,donepezil hydrochloride group was given donepezil hydrochloride by 0.45 mg/(kg·d).After four weeks of continuous gastric gavage,the rat body weight,fasting insulin(FINS),fasting blood glucose(FBG),serum total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C),and triglycerides(TG)were measured,and the behavioral changes of rats and the pathological changes of hippocampal neurons were observed.Results Compared with the diabetes model group,the weight of the rats in the high-dose Chinese medicine group was controlled(P<0.05),FBG,TG,and LDL-C were significantly reduced(P<0.01,P<0.05);the rats’escape latency was significant shortened(P<0.05),and the number of crossing platforms increased(P<0.05);pathology suggested that the nucleus pyknosis was decreased,the number of degenerated necrotic cells decreased,and the number of Nissl bodies increased.Conclusion Yizhi Jiedu Decoction can significantly improve the glucose and lipid metabolism disorder,promote the secretion of insulin,and improve the cognitive dysfunction of diabetic rats.
