血管内皮生长因子及其基因多态性与脑微出血的关系 被引量:1

Relationship between Vascular Endothelial Growth Factor and Its Gene Polymorphism and Cerebral Microbleeds

摘要 目的:探讨血管内皮生长因子(VEGF)及其基因多态性与脑微出血的关系,为脑微出血的诊治提供新思路。方法:纳入脑微出血患者76例作为病例组,同期无脑微出血患者75例作为对照组,采用酶联免疫吸附法测定血清VEGF浓度,应用聚合酶链式反应—限制性片段长度多态性(PCR-RFLP)技术检测VEGF-2578C/A基因多态性,比较两组患者血清VEGF浓度及VEGF-2578C/A基因型分布。结果:(1)病例组血清VEGF浓度显著高于对照组[分别为(172.98±137.31)pg/ml、(89.19±19.50)pg/ml],两组比较有统计学差异(P<0.05)。(2)多因素分析显示,年龄、高血压为脑微出血发生的危险因素,VEGF是脑微出血发生的独立危险因素。(3)VEGF-2578C/A基因型(CC、CA和AA)与等位基因的分布在病例组和对照组差异有统计学意义(P<0.05),且CC基因型脑微出血的发生风险是AA基因型的4.714倍。结论:VEGF水平升高与脑微出血的发生相关。VEGF-2578C/A基因多态性与脑微出血相关,CC基因型为易感基因,携带C等位基因很可能促进脑微出血的发生。 Objective:To explore the relationship between vascular endothelial growth factor(VEGF)and its gene polymorphism and cerebral microhemorrhage,and to provide a new idea for the diagnosis and treatment of cerebral microbleeds.Methods:76 patients with cerebral microbleeds were selected as case group and 75 patients without cerebral microbleeds as control group.Serum VEGF concentration was measured by enzyme linked immunosorbent assay(ELISA)and VEGF-2578C/A gene polymorphism was detected by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).The serum VEGF concentration and VEGF-2578C/A genotype distribution were compared between the two groups.Results:(1)The serum VEGF concentration in the case group was significantly higher than that in the control group[(172.98±137.31)pg/ml,(89.19±19.50)pg/ml,respectively].(2)Multivariate analysis showed that age and hypertension were the risk factors of cerebral microbleeds,and VEGF was the independent risk factor of cerebral microbleeds.(3)The distribution of VEGF-2578C/A genotypes(CC,CA and AA)and alleles in case group was significantly different from that in control group,and the risk of cerebral microbleeds with CC genotype was 4.714 times higher than that of AA genotype.Conclusion:The increase of VEGF level is related to the occurrence of cerebral microbleeds.VEGF-2578C/A gene polymorphism is associated with cerebral microbleeds.CC genotype is a susceptible gene,and carrying C allele may promote the occurrence of cerebral microbleeds.
