摘要
为了探讨DIRAS3基因在附植期猪子宫内膜组织中的表达情况及其对胚胎附植数的影响,试验采用实时荧光定量PCR和免疫组织化学方法分析了DIRAS3基因在妊娠第15,26,50天大白猪子宫内膜组织中的表达和定位情况,并通过小鼠腹腔注射试验检测DIRAS3基因对胚胎附植数的影响。结果表明:妊娠第15天大白猪子宫内膜组织中DIRAS3基因的相对表达量极显著高于妊娠第26,50天(P<0.01),且阳性信号主要集中在子宫内膜腔上皮细胞中,在子宫腺体上皮细胞中有零星分布;而在妊娠第26,50天中,子宫内膜腔上皮细胞中均未观察到明显阳性信号,仅在子宫腺体上皮细胞中存在零星分布。小鼠腹腔注射DIRAS3单克隆抗体后,小鼠胚胎附植数显著减少(P<0.05)。说明妊娠过程中DIRAS3基因在附植期子宫内膜中呈现高水平表达,且可能参与胚胎附植过程的调控。
In order to investigate the expression of DIRAS3 gene in porcine endometrium during implantation and its influence on the number of embryo implantation, real-time fluorescent quantitative PCR and immunohistochemistry methods were used to analyze the expression and localization of DIRAS3 gene in endometrium tissues of Yorkshire on days 15, 26 and 50 of pregnancy. The effect of DIRAS3 gene on the number of embryo implantation was detected by intraperitoneal injection in mice. The results showed that the DIRAS3 expression level were extremely significant higher on day 15 of pregnancy compared with day 26 and 50 of pregnancy(P<0.01) in endometrial tissue, and the positive signals were mainly concentrated in the endometrial luminal epithelial cells and scattered in the glandular epithelial cells. However, on the day 26 and 50 of pregnancy, no obvious positive signal was observed in the endometrial luminal epithelial cells, only scattered in the glandular epithelial cells. After intraperitoneal injection of DIRAS3 monoclonal antibody, the number of embryo implantation was decreased significantly in mice(P<0.05). It indicated that DIRAS3 was highly expressed in the endometrium during implantation and might be involved in the regulation of embryo implantation.
