胆酸对代谢性炎症模型大鼠糖及脂代谢的影响 被引量:3

摘要 为探讨胆酸对代谢性炎症模型大鼠糖及脂代谢的影响,试验用饲喂60 d高脂饲料的48只SD大鼠建立代谢性炎症模型,均分为模型组和胆酸高(150 mg/kg)、中(50 mg/kg)、低剂量(16.7 mg/kg)组,连续灌胃给药30 d,并以饲喂基础饲料的12只SD大鼠作为阴性对照组,分别在停药第1天、第3天检测葡萄糖耐量及胰岛素耐量,停药第5天测定天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、碱性磷酸酶(ALP)活性,白蛋白(ALB)、总蛋白(TP)、总胆酸(TBA)、胆固醇(CHO)、高密度脂蛋白胆固醇(HDL)、低密度脂蛋白胆固醇(LDL)、三酰甘油(TG)、游离脂肪酸(FFA)含量,计算大鼠增重率,测定脂肪湿重(WAT)并计算脂肪系数。结果表明:与阴性对照组相比,模型组增重率、脂肪系数、WAT、TG含量极显著升高(P<0.01)、ALP活性显著升高(P<0.05);与模型组相比,胆酸高剂量组增重率、WAT、FFA、TG含量极显著降低(P<0.01),胆酸中、低剂量组的ALP活性及FFA含量显著降低(P<0.05),胆酸中剂量组的增重率显著降低(P<0.05)。给予葡萄糖120分钟时,模型组血糖值较阴性对照组显著上升(P<0.05),而较胆酸高、中、低剂量组极显著降低(P<0.01)。腹腔内注射胰岛素后120分钟时,模型组血糖值较阴性对照组显著上升(P<0.05),胆酸高剂量组血糖值较模型组显著降低(P<0.05)。说明高剂量胆酸对代谢性炎症模型大鼠起到降脂、抑制体重增长并改善胰岛素敏感性的作用。 To investigate the effects of cholic acid on glucose and lipid metabolism in rats with metaflammation model, 48 SD rats fed with high-fat diet for 60 days were used to establish a metabolic inflammation model, and were divided into five groups: model group(MOD), high-cholic acid group(CH), medium-cholic acid group(CM), and low-dose cholic acid group(CL). Each group of 12 rats was given continuous intragastric administration for 30 days. SD rats fed with basic diet were used as negative control and the glucose tolerance and insulin tolerance were detected on the first and third days after drug withdrawal. On the fifth day after drug withdrawal, the activities of aspartate aminotransferase(AST), alanine aminotransferase(ALT), alkaline phosphatase(ALP) were determined. Albumin(ALB), total protein(TP), total cholic acid(TBA), cholesterol(CHO), high density lipoprotein(HDL), low density lipoprotein(LDL), triglyceride(TG), free fatty acid(FFA) content were measured, and the weight gain rate of rats was calculated, and the wet weight of fat(WAT) was measured and the fat coefficient was calculated. The results showed that compared with the NC group, the weight gain rate, fat coefficient, WAT and TG content in MOD group were significant increased(P<0.01), and the ALP activity was significantly increased in MOD group(P<0.05). Compared with MOD group, the weight gain rate, WAT, FFA and TG content of the CH group were extremely significant decreased(P<0.01), the ALP activity and FFA content of the CM and CL group were significantly decreased(P<0.05), and the weight gain rate of the CM group was significantly decreased(P<0.05). At 120 min after glucose administration, the blood glucose value of MOD was significantly higher than that of the NC group(P<0.05), but was extremely significant lower than that of the CL group(P<0.01). At 120 min after intraperitoneal injection of insulin, the blood glucose value of MOD group was significantly higher than that of NC group(P<0.05), while that of CH group was significantly decreased compared with NC group(P<0.05). It indicated that high dose of cholic acid could reduce blood fat, inhibit weight gain and improve insulin sensitivity in rats with metaflammation model.
