龙葵银消浸膏灌胃给药BALB/c小鼠14天的毒性评价

Toxicity Evaluation of Longkui Yinxiao Extract(龙葵银消浸膏)by Intragastric Administration for 14 days in BALB/c Mice

目的:探究龙葵银消浸膏毒性,为临床用药安全性提供参考。方法:20只BALB/c小鼠按体质量随机分为空白组和给药组,雌雄各半,每组10只。给药组小鼠给药剂量为0.75 g/mL,给药体积0.5 mL,空白组小鼠给予等体积生理盐水,1次/d,连续灌胃给药14 d,每日记录小鼠体质量,末次给药后取血检测血清肝肾功能生化指标,测定肝脏、心脏、脾脏、肺脏、肾脏质量及脏器系数,并检测脏器病理组织学改变。结果:给药组小鼠体质量、脾脏质量及脾脏系数均明显低于空白组(P<0.05);给药组小鼠血清CREA水平低于空白组(P<0.05),血清BUN、UA、AST、ALT水平与空白组比较,差异无统计学意义(P>0.05);龙葵银消浸膏能够引起小鼠肝脏炎症反应,肾脏肾间质轻微淤血。结论:龙葵银消浸膏在最大给药剂量连续给药14 d对小鼠肝肾可能具有潜在毒性。

Objective:To clarify the toxicity of Longkui Yinxiao Extract and provide beneficial exploration for clinical medication.Methods:Totally 20 BALB/c mice were randomly divided into blank group and Longkui Yinxiao Extract group,10 rats in each group,half male and half female.The Longkui Yinxiao Extract group was given a maximum dose of 0.75 g/mL for 14 consecutive days intragastrically,and the volume was 0.5 mL.The blank group was given the same amount of normal saline.After the last dose,the indexes of liver and kidney function were measured.The weight of liver,heart,spleen and kidney were measured,and the coefficients of organs were calculated.The histopathological changes of the organs were detected.Results:Longkui Yinxiao Extract group showed lower body weight,spleen weight and spleen coefficient than blank group(P<0.05).Longkui Yinxiao Extract group showed lower serum CREA than blank group(P<0.05).There was no significant difference in serum BUN,UA,AST and ALT levels between Longkui Yinxiao Extract group and blank group(P>0.05).Histopathological examination revealed that Longkui Yinxiao Extract can cause liver secretion reaction and renal interstitial micro congestion.Conclusion:The maximum dose of Longkui Yinxiao Extract may has potential toxicity to liver and kidney by intragastric administration for 14 days in BALB/c mice.
