面部色素性疾病中药面膜专利处方分析及作用机制探讨 被引量:3

Analysis of the Prescription Ingredients and Mechanism of Action of the Patented Traditional Chinese Medicine Mask for Facial Pigmented Diseases

摘要 目的采用数据挖掘及网络药理学方法分析面部色素性疾病的中药面膜专利处方的成分作用机制及有效靶点。方法利用国家知识产权局的专利公布公告平台统计近20年中药面膜专利数量,运用R语言对中药进行频次和关联规则分析,获得其中的核心药物,于中药系统药理学分析平台数据库(TCMSP)对其进行成分、靶点筛选,将所得靶点与疾病数据库GeneCards、OMIM、DisGeNET、DrugBank中筛选得到的疾病靶点进行匹配,获取核心药组治疗面部色素性疾病的关键靶点;采用STRING平台、DAVID数据库对关键靶点进行蛋白互作网络(PPI);最后通过Cytoscape3.8.0构建中药-成分-关键靶点-通路网络。结果共检索纳入符合标准的中药面膜专利433条,分析出核心药物6味(白及、白术、白芷、茯苓、当归、珍珠),这些药物已探明的有效成分有β-谷固醇、豆甾醇、茯苓酸等共66个,关联的疾病靶点1475个,疾病与核心药组的共同靶点23个,涉及癌症信号通路、肿瘤坏死因子信号通路、细胞凋亡等相关通路30条。结论面部色素性疾病的中药应用可多考虑补益药与其他药物的配伍,这类处方以三萜类和香豆素类化合物为主要有效成分,以HSP90AA1、PTPN1、EGFR、CASP3等为重要靶点,通过癌症信号通路、细胞凋亡、p53等通路发挥抗炎、抑制酪氨酸酶活性,减少黑素的生成。 Objective Data mining and network pharmacology methods were used to analyze the mechanism of action and effective targets of the patent prescription of traditional Chinese medicine mask for facial pigmentation diseases.Methods The patent announcement platform of the State Intellectual Property Office was used to count the number of TCM facial mask patents in recent 20 years.R language was used to analyze the frequency and association rules of traditional Chinese medicine,and the core drugs were obtained.The constituents and targets were screened in TCMSP database.The obtained targets were matched with the disease targets screened in the disease databases GeneCards,OMIM,Disgenet and DRUGBANK to obtain the key targets of the core drug group for the treatment of facial pigmentation diseases;Protein interaction network(PPI)for key targets using String platform and David database.Finally,the network of TCM-component-key target-pathway was constructed by Cytoscape3.8.0.Results Patent retrieval into accord with standard of traditional Chinese medicine facial mask 433 article,analyzing the core drugs 6 flavour(white atractylodes,angelica dehurica,and,poria cocos,angelica,pearl),these drugs have proven effective components have a beta valley sterol,stigmasterol,poria cocos acid,etc.A total of 66,targets 1475 associated diseases,diseases with the core medicine group of 23 common targets,It involved 30 cancer signaling pathways,tumor necrosis factor signaling pathways,apoptosis and other related pathways.Conclusion In the application of traditional Chinese medicine for facial pigment diseases,the compatibility of tonic drugs and other drugs can be considered.In this kind of prescription,trterpenoids and coumarins are the main active ingredients,and Hsp90aa1,PTPN1,EGFR and Casp3 are the important targets,and anti-inflammatory and tyrosinase inhibition activities can be played through cancer signaling pathways,cell apoptosis,p53 and other pathways.Reduces melanin production.
