阿霉素联合姜黄素抑制脑胶质瘤细胞增殖研究

摘要 本实验采取体外培养脑神经胶质瘤U251细胞株的方法,分别选用阿霉素和姜黄素对其进行药效实验。CCK-8实验检测不同浓度阿霉素、姜黄素和两药联合作用细胞48 h的抑制率。Western blot实验检测抑癌基因P53蛋白的表达。实验发现阿霉素和姜黄素均可抑制U251细胞的增殖,两药联用可提高抑制率,其机制可能与促进P53蛋白的表达有关。 In this experiment,the brainglioma U251 cell line was cultured in vitro,adriamycin and curcumin were used to test the efficacy of doxorubicin and curcumin respectively.CCK-8 assay was used to detect the inhibition rate of cells treated with different concentrations of doxorubicin,curcumin and the combination of the two drugs for 48 hours.The expression of tumor suppressor gene P53 protein was detected by Western blot assay.It was found that both adriamycin and curcumin could inhibit the proliferation of U251 cells,and the combination of doxorubicin and curcumin could increase the inhibition rate,The mechanism may be related to the promotion of P53 protein expression.
