太赫兹波治疗对血管性痴呆大鼠学习记忆及CD34、VEGF蛋白表达的影响 被引量:3

Effects of terahertz magnetic wave therapy on learning and memory,CD34 and VEGF protein expression in vascular dementia rats

摘要 目的:观察太赫兹波治疗对血管性痴呆(VD)大鼠学习记忆及相关蛋白表达的影响。方法:采用改良后双侧颈总动脉永久性结扎法(2-VO)法制备模型,将SD大鼠随机分成假手术组,模型组,西药组(0.01g/kg尼莫地平),低、中、高太赫兹波组,通过Morris水迷宫检测神经行为学改变,HE染色观察病理变化情况,免疫组化检测脑皮质及胼胝体中CD34、血管内皮生长因子(VEGF)蛋白表达,CD34标记检测微血管密度(MVD)。结果:模型组较假手术组学习记忆明显障碍(P<0.05),皮质、胼胝体中CD34及VEGF蛋白表达增强,MVD显著增多(P<0.05)。与模型组比较,太赫兹波组VD大鼠学习能力显著提高(P<0.05),皮质、胼胝体中CD34及VEGF蛋白表达增强,MVD显著增多(P<0.05)。与西药组比较,中太赫兹波组CD34及VEGF蛋白表达增强,MVD显著增多(P<0.05)。结论:太赫兹波治疗可改善VD大鼠学习记忆能力,其机制可能与增强VD大鼠脑组织中CD34、VEGF蛋白表达,增多MVD,促血管新生,缓解大脑缺血状态有关。 Objective:To observe the effects of terahertz magnetic wave therapy instrument on learning memory and related protein expression in vascular dementia(VD)rats.Methods:The 2-VO method was used to prepare models,and SD rats were randomly divided into sham operation group,model group,western medicine group(0.01g/kg nimodipine),low,medium and high terahertz magnetic wave group.Morris water maze was used to detect neurobehavior the pathological changes were observed by HE staining,the expression of CD34 and VEGF protein in the cerebral cortex and corpus callosum were detected by immunohistochemistry,and MVD was detected by CD34 labeling.Results:Compared with the sham operation group,the model group’s the learning and memory were significantly impaired(P<0.05),the expression of CD34 and VEGF protein was increased,and the MVD was increased(P<0.05).Compared with the model group,the terahertz magnetic wave group’s learning ability was significantly improved(P<0.05),the expression of CD34 and VEGF protein in cortex and corpus callosum increased,and MVD increased(P<0.05).Compared with the western medicine group,the CD34 and VEGF protein expression in the medium terahertz magnetic wave group was increased,and the MVD increased(P<0.05).Conclusion:Terahertz magnetic wave treatment can improve the learning and memory ability of VD rats.The mechanism may be related to enhancing the expression of CD34 and VEGF protein in the brain tissue of VD rats,increasing MVD,promoting angiogenesis,and alleviating cerebral ischemia.
