仙茅不同炮制品抗维甲酸致雄性大鼠骨质疏松症作用机制研究 被引量:5

Study on the mechanism of different processed products of Rhizoma Curculigins on osteoporosis male rats induced by retinoic acid

摘要 目的:研究仙茅不同炮制品抗维甲酸致雄性大鼠骨质疏松症的作用机制。方法:以仙灵骨葆片药液为阳性组,采用酶联免疫吸附法(ELISA)检测了仙茅不同炮制品组灌胃水提液4周后维甲酸致骨质疏松症雄性大鼠血清抗酒石酸盐酸性磷酸酶(TRACP5B)水平和作用机制指标血清护骨素(OPG)、血钙(Ca)和磷(P)、白细胞介素-6(IL-6)、肿瘤坏死因子(TNF-α)、白细胞介素-1β(IL-1β)、基质金属蛋白酶-9(MMP-9)、基质金属蛋白酶抑制因子-1(TIMP-1)、核因子κB(NF-κB)受体活化因子配基(RANKL)的水平,采用一步法逆转录聚合酶链反应方法(RT-PCR)测定大鼠肝脏铁调素mRNA表达水平。结果:仙茅各炮制品水提液对维甲酸致骨质疏松症大鼠的作用机制指标均有不同程度纠正作用,并且均能下调肝脏铁调素mRNA表达水平,以盐仙茅、酒仙茅效果较好,姜仙茅、萸仙茅及生品次之。结论:仙茅各炮制品均能有效对抗维甲酸致骨质疏松症,其中酒仙茅、盐仙茅抗骨质疏松效果较为显著。 Objective:To study the mechanism of anti-osteoporosis of different processed products of Rhizoma Curculigins.Methods:With the Xianling Gubao Tablets as the positive control group,all decoctions concentration of different products for 4 weeks were compared to study the content of TRACP5B,OPG,Ca,P,IL-6,TNF-α,IL-1β,MMP-9,TIMP-1,RANKL in serum of normal and retinoic acid induced osteoporosis rats by ELISA,and the expression of hepcidin mRNA in rat liver was determined by RT-PCR.Results:The raw and its processed products of Rhizoma Curculigins have a certain degree of corrective action for the mechanism indexes of osteoporosis rats,and can cut hepcidin mRNA expression level.The effects of stirfrying with salt-water and stir-frying with wine are better,stir-frying with ginger,stir-frying with fructus evodiae and crude take second place.Conclusion:The raw and its processed products of Rhizoma Curculigins are effective against retinoic acid induced osteoporosis,curative effect of stir-frying with salt-water and stir-frying with wine are better.
