LINC00511过表达促进宫颈癌HeLa细胞的增殖、迁移和侵袭

The overexpression of LINC00511 promotes the proliferation,migration and invasion of HeLa cells

目的:研究长链非编码RNA(LncRNA)LINC00511过表达对宫颈癌HeLa细胞增殖、迁移和侵袭能力的影响并探索其潜在机制。方法:利用荧光定量PCR(qRT-PCR)及Western blotting检测宫颈癌HeLa细胞中LINC00511 mRNA和蛋白表达;利用细胞转染试验过表达宫颈癌细胞中的LINC00511,利用CCK-8检测法、Transwell实验探究过表达LINC00511对宫颈癌HeLa细胞增殖、迁移和侵袭的影响;通过Western blotting检测HeLa细胞中EMT相关蛋白的表达情况,探究潜在作用机制。结果:HeLa细胞中LINC00511 mRNA和蛋白表达水平显著高于正常宫颈内皮细胞(P<0.001);CCK-8法及Transwell实验显示,过表达LINC00511促进了宫颈癌HeLa细胞的增殖、迁移和侵袭能力(P<0.01);Western blotting检测显示,过表达LINC00511后宫颈癌HeLa细胞中E-cadherin蛋白表达水平显著下调,而Vimentin和α-SMA蛋白表达水平显著上调(P<0.05)。结论:宫颈癌HeLa细胞中LINC00511显著高表达,可能是宫颈癌潜在的癌基因,LINC00511过表达可促进宫颈癌HeLa细胞的增殖、迁移和侵袭,可能通过调控EMT过程来调控宫颈癌细胞的生物学行为。

Objective:To study the effect of LINC00511 overexpression on the proliferation,migration and invasion of HeLa cells in cervical cancer and explore its potential mechanism.Methods:Fluorescence quantitative PCR(qRT-PCR)and Western blotting were used to detect the expression of LINC00511 mRNA and proteins in HeLa of cervical cancer cells.Using cell transfection test to overexpress LINC00511 in cervical cancer cells,the effects of overexpression LINC00511 on the proliferation,migration and invasion of HeLa in cervical cancer cells were investigated by CCK-8 assay and Transwell assay.Western blotting was used to detect the expression of EMT-related proteins in HeLa cells to explore the potential mechanism of action.Results:The expression levels of LINC00511 mRNA and protein in HeLa cells were significantly higher than that in normal cervical endothelial cells(P<0.001).CCK-8 and Transwell assay showed that overexpression of LINC00511 promoted the proliferation,migration and invasion of HeLa in cervical cancer cells(P<0.01).Western blotting showed that the expression level of E-cadherin in HeLa of cervical cancer cells was significantly down-regulated after LINC00511 overexpression,while the expression levels of Vimentin andα-SMA were significantly up-regulated(P<0.05).Conclusion:The significantly high expression of LINC00511 in HeLa of cervical cancer cells may be a potential oncogene of cervical cancer.The overexpression of LINC00511 can promote the proliferation,migration and invasion of HeLa of cervical cancer cells,and may regulate the biological behavior of cervical cancer cells by regulating the EMT process.
