摘要
目的:通过数据挖掘分析E2F7在肾透明细胞癌(clear cell renal cell carcinoma,ccRCC)组织中的表达特征及临床意义。方法:基于GEPIA、LinkedOmics数据库分析E2F7在ccRCC中的差异性表达及其与患者预后的关系。应用WebGestalt工具分析与E2F7表达相关的基因。结果:E2F7 mRNA在ccRCC组织中表达水平高于正常肾组织。Linkedomics数据库分析显示E2F7 mRNA在ccRCC病理分期中存在表达差异(P<0.05),且随着病理分期的升高,表达水平有上调趋势。E2F7 mRNA在不同T、M、N分期(P<0.05)中存在差异性表达。生存分析表明,E2F7的表达水平与ccRCC患者的总体生存率(Logrank P=0.023)与无复发生存率(Logrank P=0.041)显著相关,此外,E2F7表达水平与BUB1B、CDC6、CCNE2、CCNE1(r分别为0.8277、0.7204、0.6778、0.6125,P均<0.01)基因呈显著正相关,与PINK1、KAT5、HSD17B8(r分别为-0.5110、-0.5084、-0.5029,P均<0.01)基因呈显著负相关。E2F7表达相关基因富集分析与DNA复制、细胞周期以及NF-kappa B信号通路(NF-kappa B signaling pathway)相关。结论:在ccRCC中E2F7的表达与患者的病理分期、T分期、N分期、M分期及预后有一定的相关性,因此E2F7可能成为ccRCC的预后标志物及潜在治疗靶点。
Objective:To analyze the expression characteristics and clinical significance of E2F7 in clear cell renal cell carcinoma based on TCGA database.Methods:The differential expression of E2F7 and the association between E2F7 and clinical prognosis in clear cell renal cell carcinoma were analyzed based on the GEPIA and LinkedOmics databases.The genes related to the expression of E2F7 were analyzed with WebGestalt tool.Results:The analysis based on the GEPIA showed that the expression level of E2F7 mRNA in ccRCC tissues was higher than that in normal renal tissues.The Linkedomics database analysis showed that E2F7 mRNA was differentially expressed in the pathological stage of clear cell renal cell carcinoma(P<0.05),and the expression level tended to be up-regulated with the increase of the pathological stage.E2F7 mRNA was differentially expressed in different T,M and N stages(P<0.05).Survival analysis showed that E2F7 expression level was significantly associated with overall survival(Logrank P=0.023)and disease free survival(Logrank P=0.041)in ccRCC patients.In addition,the expression level of E2F7 was positively correlated with that of BUB1B,CDC6,CCNE2 and CCNE1(r values were 0.8277,0.7204,0.6778 and 0.6125,respectively,all P<0.01),and negatively correlated with those of PINK1,KAT5 and HSD17B8(r values were-0.5110,-0.5084 and-0.5029,respectively,all P<0.01).The enrichment analysis of genes associated with E2F7 expression was associated with DNA replication,cell cycle,and the NF-kappa B signaling pathway.Conclusion:The expression level of E2F7 mRNA was significantly correlated with the pathological stage,T stage,N stage,M stage and prognosis in patients with clear cell renal cell carcinoma.Therefore,E2F7 has the potential to become a prognostic marker and therapeutic target of clear cell renal cell carcinoma.
